Two new hybrid isoprenoids, named indotertine B (2) and drimentine H (3), along with the known analogue drimentine C (4), were isolated from the reed rhizosphere soil derived actinomycete Streptomyces sp. CHQ-64. The structures of these compounds, including absolute configurations, were elucidated by extensive NMR, MS, and CD analyses. Indotertine B (2) exists as a pair of rotamers about the N-C(O) bond with a 2:1 ratio and displays activities against HCT-8 and A549 tumor cell lines with IC50 values of 6.96 and 4.88 mu M, respectively.

Marine-Derived Natural Product HDYL-GQQ-495 Targets P62 to Inhibit Autophagy

Summary: Autophagy is involved in pathophysiological processes and considered as a potential target for drug discovery. We used a cell line with fluorescent protein to screen marine compounds and identified HDYL-GQQ-495 as a potential autophagy modulator. Further experiments showed that HDYL-GQQ-495 inhibited autophagy and induced cell death.

Biotransformation of Penindolone, an Influenza A Virus Inhibitor

Summary: In this study, the metabolic route of PND in rats was revealed using UHPLC-HRMS. A total of 25 metabolites were identified in rat bile, feces, and urine, and the metabolic pathways were proposed. It was found that PND was mainly eliminated through glucuronide conjugation in bile and excreted in feces, with significant species differences in the metabolism of PND in rat, mouse, and human liver microsomes.

Stictamycin, an Aromatic Polyketide Antibiotic Isolated from a New Zealand Lichen-Sourced Streptomyces Species

Summary: This article describes the isolation and characterization of a new aromatic polyketide, stictamycin, which has activity against Staphylococcus aureus. Stictamycin was identified from organic extracts of Streptomyces sp. 438-3, an isolate from the New Zealand lichen Sticta felix, through metabolic profiling and bioactivity guided fractionation. NMR analyses determined the planar structure and relative configurations of stereo centers of stictamycin, and the absolute configuration was elucidated through comparison of experimental and theoretical ECD spectra. Whole-genome sequencing and biosynthetic gene cluster (BGC) analysis revealed an atypical type II polyketide (T2PKS) BGC capable of assembling polycyclic-aromatic ring skeletons in Streptomyces sp. 438-3, which was confirmed to be responsible for stictamycin biosynthesis through cloning and knockout studies.

Overexpression of Global Regulator SCrp Leads to the Discovery of New Angucyclines in Streptomyces sp. XS-16

Summary: In this study, six angucyclines, including three previously unreported compounds, were isolated from Streptomyces sp. XS-16 by overexpressing the native global regulator of SCrp. The structures were characterized using NMR and spectrometry analysis, as well as assisted by ECD calculations. All compounds were evaluated for their antitumor and antimicrobial activities, and compound 1 showed varying inhibitory activities against different tumor cell lines with IC50 values ranging from 0.32 to 5.33 μM.

Design, synthesis, and biological evaluation of novel penindolone derivatives as potential inhibitors of hemagglutinin-mediated membrane fusion

Summary: The development of anti-influenza drugs with novel mechanisms is crucial in combating the ongoing threat of influenza A virus (IAV). Hemagglutinin (HA) has been identified as a potential target for IAV therapy. In this study, 65 derivatives of penindolone (PND) were designed and synthesized to enhance bioactivity and understand the structure-activity relationships (SARs). Compound 5g showed high affinity to HA and exhibited stronger inhibition of HA-mediated membrane fusion compared to PND. Additionally, oral administration of 5g demonstrated better anti-IAV effects in terms of reducing virus titer, attenuating weight loss, and improving survival in IAV-infected mice.

Activation of a Silent Polyketide Synthase SlPKS4 Encoding the C7-Methylated Isocoumarin in a Marine-Derived Fungus Simplicillium lamellicola HDN13-430

Summary: This study reports the activation of a silent nonreducing polyketide synthase that encodes a C-7-methylated isocoumarin and the elucidation of its enzymatic mechanism in Simplicillium lamellicola HDN13-430. Additionally, feeding experiments revealed the host enzymes can convert the product into its hydroxylated derivatives, and these compounds showed moderate radical scavenging activities.

Enhanced Phycocyanobilin Production in Escherichia coli by Fusion-Expression of Apo-Proteins with Signal Peptides

Summary: This study used a fusion-expression strategy to improve the production of phycocyanobilin (PCB) in Escherichia coli. The results showed that by fusing apo-proteins with signal peptides, the secretion of phycocyanin was refined and the production of PCB was successfully enhanced by 3.7-fold.

Bio-inspired construction of a tetracyclic ring system with an avarane skeleton: total synthesis of dactyloquinone A

Summary: The asymmetric synthesis of marine meroterpenoid dactyloquinone A was achieved for the first time. A bio-inspired reaction was used to construct the key intermediate, a 6/6/6/6 tetracyclic core derivative, which facilitated the migration of methyl groups from the aureane skeleton to the avarane skeleton. This study lays the groundwork for further synthetic exploration of this family of natural products.

Linear polyketides produced by co-culture of Penicillium crustosum and Penicillium fellutanum

Summary: Two new polyketides, penifellutins A and B, were isolated from a co-culture of deep-sea-derived fungi, with difficulties in determining their configurations due to chiral central crowdedness. Compounds 1 and 2 showed inhibitory activity on liver hyperplasia of zebrafish larvae, while compounds 3 and 4 did not exhibit any activity, indicating the importance of two carboxyls as active sites in the structure.

Structural diversity and biological activity of natural p-terphenyls
