Cytotoxic Naphthoquinones from Alkanna cappadocica

In a continuing program to discover new anticancer agents from plants, especially naphthoquinones from the Alkanna genus, Alkanna cappadocica was investigated. Bioassay-guided fractionation of a dichloromethane/methanol (1:1) extract of the roots led to the isolation of four new and four known naphthoquinones. The known compounds are 11-deoxyalkannin (1), beta,beta-dimethylacrylalkannin (2), 11-O-acetylalkannin (3), and alkannin (4). The new compounds 5-O-methyl-11-deoxyalkannin (5), 8-O-methyl-11-deoxyalkannin (6), 5-O-methyl-11-O-acetylalkannin (7), and 5-O-methyl-beta,beta-dimethylacrylalkannin (8) were characterized by spectroscopic analyses (LC-ESIMS, 1D and 2D NMR). Cytotoxicity of the isolated compounds was evaluated versus 12 human cancer cell lines, HT-29, MDA-MB-231, PC-3, AU565, Hep G2, LNCaP, MCF7, HeLa, SK-BR-3, DU 145, Saos-2, and Hep 3B together with two normal cell lines, VERO and 3T3, by using the MIT assay. Compound 7 showed remarkable cytotoxicity with IC50 values between 0.09 and 14.07 mu M. It was more potent than the other compounds in six out of 12 cancer cell lines and the positive controls doxorubicin and etoposide. The mono-O-methylated alkannin derivatives and their cytotoxicities are reported for the first time.

Benzodiazepine Derivatives from Marine-Derived Streptomyces cacaoi 14CM034

Summary: A new natural product with pyrrolobenzodiazepine (PBD) framework, 7-methoxy-8-hydroxy cycloanthranilylproline (2), was isolated from marine-derived actinobacterium Streptomyces cacaoi 14CM034, along with cycloanthranilylproline (1). The compounds were structurally elucidated using various analytical techniques and showed notable antimicrobial activity. This study demonstrated the presence of PBD derivatives in S. cacaoi for the first time.

Undescribed polyether ionophores from Streptomyces cacaoi and their antibacterial and antiproliferative activities

Summary: Polyether ionophores are a group of naturally occurring compounds mainly produced by Streptomyces species with various bioactivities. In this study, four undescribed and nine known polyether compounds were identified from Streptomyces cacaoi. The antimicrobial activity assays showed strong inhibitory effects of these compounds, including an undescribed compound, on the growth of Bacillus subtilis and methicillin-resistant Staphylococcus aureus (MRSA). Additionally, two compounds displayed significant cytotoxicity, with the C15-demethoxy derivative showing higher selectivity towards cancer cells than K41-A. Mechanistic experiments revealed that the uninvestigated compound acts as a multitarget compound, inhibiting autophagic flux, inducing reactive oxygen species formation, abolishing proteasome activity, and stimulating ER stress.

Characterization of different chitosanases of Bacillus strains and their application in chitooligosaccharides production

Is telomerase a hidden player? Therapeutic potential of natural telomerase activators against age-related diseases

Summary: There is a high demand for novel approaches to treat age-related diseases, which are a major cause of death worldwide. Dysfunctional telomeres, which accelerate the aging process, are associated with many of these diseases. Telomerase, an enzyme responsible for maintaining healthy telomeres, has shown promise as a target for treatment. Natural products containing telomerase activators have protective and therapeutic effects against age-related diseases, particularly neurodegenerative, cardiovascular, and osteodegenerative disorders. However, the role of telomerase activation is often overlooked in studies.

The Regulation of Cyclins and Cyclin-Dependent Kinases in the Development of Gastric Cancer

Summary: Gastric cancer is characterized by uncontrolled growth and metastasis of gastric epithelial cells, which is predominantly in adenocarcinoma form. The regulation of gastric cell growth is influenced by key cell cycle regulators, including Cyclins and Cyclin-dependent kinases (CDKs). Dysregulation of these molecules can lead to disturbances in cell cycle dynamics and the development of gastric cancer. Numerous studies have examined the roles of Cyclins and CDKs in gastric cancer, and this review aims to compile evidence of upstream regulators that activate or inhibit Cyclins and CDKs. It serves as a comprehensive resource for future studies and hypothesis generation.

neo-Clerodanes from Teucrium divaricatum subsp. divaricatum and their biological activity assessment

Summary: Fifteen new neo-clerodane diterpenoids, including two undescribed glycosides, as well as a known iridoid glycoside and a phenylpropanoid glycoside, were isolated from the whole plant of Teucrium divaricatum subsp. divaricatum. Their structures were determined through spectral data analysis, and their anti-inflammatory and antimicrobial activities were evaluated. The compounds showed no significant antimicrobial activity against various bacterial and fungal strains, and were inactive up to the highest tested concentration of 50 μM on iNOS inhibitory activity.

Astragalus Saponins, Astragaloside VII and Newly Synthesized Derivatives, Induce Dendritic Cell Maturation and T Cell Activation

Summary: Astragaloside VII (AST VII), a triterpenic saponin, has potential as a vaccine adjuvant due to its ability to promote a balanced Th1/Th2 immune response. However, the underlying mechanisms of its adjuvant activity have not been fully understood. This study investigated the effects of AST VII and its analogs on human whole blood cells and mouse bone marrow-derived dendritic cells (BMDCs). The results demonstrate that AST VII and its derivatives enhance pro-inflammatory responses, support dendritic cell maturation, and activate T cells in vitro.

Biomimetic peptide-conjugated membranes for developing an artificial cornea

Tissue engineering approaches, combined with peptide-modified scaffold surfaces, offer promising alternatives for the regeneration of corneal endothelium tissue.
