Bafilomycins Produced in Culture by Streptomyces spp. Isolated from Marine Habitats Are Potent Inhibitors of Autophagy

Five new bafilomycins. F (1) to J (5), have been isolated from laboratory Cultures of two Streptomyces spp. obtained front marine sediments collected in British Columbia, and their Structures have been elucidated by detailed analysis of spectroscopic data and the synthesis of model compounds. The new bafilomycins F (1), G (2), H (3), and J (5) alone with several co-occurring known analogues showed potent inhibition of autophagy in microscopy and biochemical assays. The thiomorpholinone fragment present in bafilomycin F (1) has not previously been found in a natural product.

Summary: Studies have shown that certain sesquiterpene lactones isolated from the indigenous zoanthid Palythoa aff. clavata collected at the coast of Lanzarote, Canay Islands exhibit anti-amoebic effects, with 4-epi-arbusculin A and isobadgerin showing promising activity and low cytotoxicity. The mode of action of compounds 2 and 11 on Acanthamoeba castellanii Neff involves induction of PDC mechanisms.

Apoptosis-like cell death upon kinetoplastid induction by compounds isolated from the brown algae Dictyota spiralis

Summary: The study found that the cyclooctapeptides squamins C-F from Annona globiflora exhibit anti-amoeboid activity against Acanthamoeba, especially sensitive to A. castellanii Neff. These peptides can induce programmed cell death in Acanthamoeba, causing mitochondrial damage and increased production of reactive oxygen species (ROS), but they have no cytotoxic effects on murine macrophages.

Alciporin, a pore-forming protein as complementary defense mechanism in Millepora alcicornis

Summary: Millepora alcicornis, also known as fire coral, is a tropical hydrozoa species that produces thermolabile proteins, including phospholipase A2 (PLA2) and alciporin, a pore-forming protein (PFP). The venom contained in the nematocysts of this species displays hemolytic activity primarily through the action of alciporin, which targets sphingomyelin. Atomic force microscopy (AFM) was used to study the evolution of hemolytic damage at the nanoscale.

Metabolic Hepatic Disorders Caused by Ciguatoxins in Goldfish (Carassius auratus)

Summary: Ciguatera poisoning (CP) is a well-known foodborne disease, but little research has been done on the effects of ciguatoxins (CTXs) on fish metabolism. This study examined the changes in hepatic compounds in goldfish (Carassius auratus) fed C-CTX1 using NMR-based metabolomics. The results showed that the fish fed toxic flesh had increased liver size and altered concentrations of alanine, lactate, taurine, glucose, and glycogen. These changes may be linked to an increase in ammonium ion production.

Laurequinone, a Lead Compound against Leishmania

Summary: In this study, it was found that five compounds isolated from the red alga Laurencia johnstonii exhibited leishmanicidal activity. Among them, laurequinone showed the strongest activity and was capable of inducing programmed cell death or apoptosis in the parasite. These findings highlight the potential of laurequinone as a novel therapeutic agent against kinetoplastid parasites.

Summary: Natural Products (NP) play a crucial role in the discovery of novel drugs and biotechnological applications. However, the process of NP discovery is expensive and time-consuming, mainly due to the challenges of dereplication and structure elucidation, especially for metabolites with stereogenic centers. This review highlights recent technological and instrumental advances that address these obstacles and accelerate NP discovery for biotechnological applications. It emphasizes innovative high-throughput tools and methods for bioactivity screening, NP chemical analysis, dereplication, metabolite profiling, metabolomics, genome sequencing and/or genomics approaches, databases, bioinformatics, chemoinformatics, and three-dimensional NP structure elucidation.

Identification and characterization of novel marine oxasqualenoid yucatecone against Naegleria fowleri

Summary: This study evaluated the in vitro activity of six oxasqualenoids obtained from the red algae Laurencia viridis against N. fowleri, as well as their cytotoxicity against murine macrophages. Yucatecone showed the highest selectivity index and induce programmed cell death in treated amoebae. The presence of a ketone at C-18 was found to be the significant structural feature for activity against N. fowleri.

Marine Meroterpenoids Isolated from Gongolaria abies-marina Induce Programmed Cell Death in Naegleria fowleri

Summary: In this study, six meroterpenoids isolated from the brown algae Gongolaria abies-marina were evaluated for their activity against N. fowleri. Three compounds, Gongolarone B (1), 6Z-1-methoxyamentadione (2), and 1-methoxyamentadione (3), showed the most activity against N. fowleri, with IC50 values ranging from 13.27±0.96μM to 21.92±1.60μM. Cystomexicone B (6) exhibited the highest selectivity index (>8.5). Moreover, all these compounds induced cellular events related to apoptosis-like programmed cell death. Therefore, G. abies-marina could be a promising source of active molecules for treating N. fowleri infections.
