Isolation and Biosynthesis of Aurachin P and 5-Nitroresorcinol from Stigmatella erecta

摘要

The isolation of aurachin P (2) from Stigmatella erecta strain Pd e32 is described. Spectroscopic data, in particular NMR data, indicate it is 1 '-hydroxyaurachin A with a 1 ' R,2 ' S,3 ' R relative configuration. In addition, a further compound, 5-nitroresorcinol (4a), was isolated and identified as a novel natural product. Feeding of C-13- and N-15-labeled precursors indicated this was synthesized solely from glucose and ammonia. To account for the labeling pattern, phloroglucinol (8) is postulated as an intermediate branching off from 3-dehydroquinate (7) in the shikimate pathway.

N2H4-H2O Enabled Umpolung Cyclization of o-Nitro Chalcones for the Construction of Quinoline N-Oxides

Summary: Umpolung is a unique strategy that converts the property of an atom into the opposite one. An efficient and general method for constructing quinoline N-oxides from ortho-nitro chalcones and hydrazine has been developed under transition-metal free conditions. The strategy shows good functional group tolerance, environmental friendliness, and mild reaction conditions, with nitrogen gas as the byproduct.

Summary: Outer membrane vesicles shed from Gram-negative bacteria play important roles in resistance gene transfer, bacterial virulence, and host cell modulation. This study reports the discovery of autofluorescent vesicles derived from non-pathogenic soil-living myxobacteria, which also exhibit light-regulated antibiotic activity. These findings provide insights into the environmental impact on bacteria-derived vesicles and have implications for future therapeutic design.

Purpurascenines A-C, Azepino-Indole Alkaloids from Cortinarius purpurascens: Isolation, Biosynthesis, and Activity Studies on the 5-HT2A Receptor

Summary: Three new azepino-indole alkaloids, named purpurascenines A-C (1-3), along with a new-to-nature 7-hydroxytryptophan (4) and two known compounds, adenosine (5) and riboflavin (6), were isolated from the fruiting bodies of Cortinarius purpurascens. The structures of 1-3 were determined using spectroscopic analysis and ECD calculations. The biosynthesis of purpurascenine A (1) was investigated, and a biosynthetic route involving a direct Pictet-Spengler reaction between alpha-keto acids and 7-hydroxytryptophan (4) was proposed. Compound 1 showed no antiproliferative or cytotoxic effects against human cancer cells and exhibited some antagonistic effects on the 5-HT-dependent 5-HT2A activation and putative constitutive activity of the 5-HT2A receptor.

Marinoterpins A-C: Rare Linear Merosesterterpenoids from Marine-Derived Actinomycete Bacteria of the Family Streptomycetaceae

Summary: The chemical examination of two undescribed marine actinobacteria has led to the discovery of three rare merosesterterpenoids with unique chemical motifs. Extensive analysis using 1D and 2D NMR, MS, and combined spectroscopic data allowed for the determination of the structures of these new compounds, while also revealing the uncommon occurrence of two five-ring ethers. Additionally, the genome scanning of one actinobacteria strain provided insight into the biosynthesis of these compounds.

Identification, Isolation, and Characterization of Medipeptins, Antimicrobial Peptides From Pseudomonas mediterranea EDOX

Xanthones and anthraquinones from the soil fungus Penicillium sp. DWS10-P-6
