Seven new lanostane-type triterpenoids, seco-coccinic acids A-F (1-6) and coccinilactone A (7), were isolated from the roots of Kadsura coccinea. Their structures were established on the basis of spectroscopic data analysis. The absolute configuration at C-24 of compound 5 was confirmed by the modified Mosher's method. The cell growth inhibitory effects of these compounds were determined in human leukemia HL-60 cells, and it was found that compounds 1, 2, 3, and 5 exhibited antiproliferative effects with GI(50) values ranging from 6.8 to 42.1 mu M.

Lathyrane and Jatrophane Diterpenoids from Euphorbia helioscopia Evaluated for Cytotoxicity against a Paclitaxel-Resistant A549 Human Lung Cancer Cell Line

Di Zhou, Fengjiao Zhang, Takashi Kikuchi, Miao Yao, Kouharu Otsuki, Gang Chen, Wei Li, Ning Li

Summary: A phytochemical investigation of Euphorbia helioscopia yielded 27 macrocyclic diterpenoids, including three previously unreported lathyrane derivatives. Three of these compounds exhibited cytotoxicity against a paclitaxel-resistant lung cancer cell line, but showed no activity against the parent cancer cell line. It was observed that compounds with benzoyloxy or nicotinoyloxy substituents at a specific position displayed enhanced cytotoxic activity.

Okanin from Coreopsis tinctoria Nutt. alleviates cognitive impairment in bilateral common carotid artery occlusion mice by regulating the miR-7/NLRP3 axis in microglia

Yan Mi, Jikai Xu, Ruijia Shi, Qingqi Meng, Libin Xu, Yeshu Liu, Tingting Guo, Di Zhou, Jingyu Liu, Wei Li, Ning Li, Yue Hou

Summary: This study investigates the effect of okanin on cognitive impairment following ischemic stroke and finds that it attenuates impairment by inhibiting microglial activation and decreasing NLRP3 inflammasome activation. The study also shows that the miR-7/NLRP3 axis plays a crucial role in mediating the beneficial effects of okanin on cerebral ischemia.

Tigliane and daphnane diterpenoids from Thymelaeaceae family: chemistry, biological activity, and potential in drug discovery

Kouharu Otsuki, Wei Li

Summary: Tigliane and daphnane diterpenoids, found in Thymelaeaceae and Euphorbiaceae plants, have diverse structures due to the presence of polyoxygenated functionalities in their polycyclic skeleton. Despite being known as toxic components, these diterpenoids have shown a wide range of biological activities, including anti-cancer, anti-HIV, and analgesic properties, making them attractive in natural product drug discovery. This review focuses on naturally occurring tigliane and daphnane diterpenoids from Thymelaeaceae plants, providing an overview of their chemical structure, distribution, isolation, structure determination, chemical synthesis, and recent findings.
