Trichodermanin A, a structurally unique diterpenoid with skeletal carbons arranged compactly in a fused 6-5-6-6 ring system, has been isolated from cultures of Trichoderma atroviride (strain no. S361), an endophytic fungus isolate in Cephalotaxus fortunei. The structure of this compound was elucidated based on extensive spectroscopic methods and confirmed by X-ray diffraction analysis.

Summary: Through a series of in vitro and in vivo experiments, it has been shown that the conversion of TACs may result from two non-enzymatic steps, possibly mediated by endogenous hydrogen sulfide. Therefore, TACs are proposed to derive from a detoxification process.

Structural characterization and hepatoprotective effects of polysaccharides from Anoectochilus zhejiangensis

Summary: In this study, two new polysaccharides, AZP-1a and AZP-1d, were extracted from Anoectochilus zhejiangensis and their structural characteristics were explored. The results showed that A. zhejiangensis polysaccharides exhibited significant hepatoprotective effects and AZP-1a and AZP-1d mainly functioned through the activation of the Nrf2 signaling pathway.

However, the mechanisms underlying palmitoylation in these processes are not well understood. This study investigated the role of palmitoylation in hippocampal synaptic transmission and fear memory formation and identified the involvement of APT1-mediated depalmitoylation in the regulation of glutamatergic synaptic plasticity.

The abundance of novel skeletons, bioactivities, and biosynthetic potentials present new opportunities for drug and agrochemicals discovery, genome mining, and enzymology exploration from Trichoderma species.

Structural characterization and antinociceptive activity of polysaccharides from Anoectochilus elatus

Neogrisemycin, a Trisulfide-Bridged Angucycline, Produced upon Expressing the Thioangucycline Biosynthetic Gene Cluster in Streptomyces albus J1074

Summary: The authors isolated neogrisemycin (1) from a recombinant Streptomyces albus J1074 strain SB4061 expressing an engineered thioangucycline (TAC) biosynthetic gene cluster (BGC). They determined the structure and absolute configuration of 1 using mass spectrometry, nuclear magnetic resonance, and single-crystal X-ray diffraction analyses. They proposed that 1, like the TACs, is derived non-enzymatically from the common epoxide (8) encoded by the tac BGC, mediated by endogenous hydrogen trisulfide.

Total lignans from Vitex negundo seeds attenuate osteoarthritis and their main component vitedoin A alleviates osteoclast differentiation by suppressing ERK/NFATc1 signaling

Summary: This study investigated the anti-osteoarthritis effects of the total lignans of Vitex negundo seeds (TOV) in osteoarthritis rats, as well as the effects of its main components VN1 and VN2 on osteoclast differentiation. TOV significantly alleviated osteoarthritis symptoms and decreased inflammatory markers. VN2 inhibited osteoclast differentiation by suppressing ERK/NFATc1 signaling and actin ring formation. These findings suggest that TOV has therapeutic potential for osteoarthritis treatment and VN2 plays a key role in its efficacy.

Mechanisms of Epichloe bromicola to Promote Plant Growth and Its Potential Application for Coix lacryma-jobi L. Cultivation

Summary: Endophytic fungi, such as Epichloe bromicola, have been found to enhance plant growth and biosynthesis of secondary metabolites. In this study, the growth-promoting effects of E. bromicola on Arabidopsis thaliana seedlings were observed, and it was found that the compound IAA and 2,3-butanediol were responsible for the effects. Furthermore, the expression of IAA-related genes was upregulated after inoculation with E. bromicola, indicating the involvement of IAA in the growth-promoting mechanism.
