Increased Expression of TRPV1 in the Cortex and Hippocampus from Patients with Mesial Temporal Lobe Epilepsy

Transient receptor potential vanilloid type-1 (TRPV1) is a ligand-gated nonselective cation channel that has been well characterized in peripheral pain pathway. Recent evidence from animal models of temporal lobe epilepsy (TLE) has supported the important role of TRPV1 in epileptogenesis. In this study, we investigated the expression and cellular distribution of TRPV1 in the temporal cortex (CTX) and hippocampus (HPC) from 26 patients with mesial TLE (MTLE) compared with 12 histologically normal samples. Reverse transcription-PCR and Western blotting revealed up-regulated mRNA and protein levels of TRPV1 in the MTLE group versus the control group. Immunohistochemistry data demonstrated that TRPV1 was mainly distributed in the cell bodies and dendrites of neurons. Double-labeled immunofluorescence further revealed that TRPV1 was localized on NeuN-positive neurons and GFAP-positive astrocytes, but not on HLA-positive microglia. In addition, its co-localization with glutamate and gamma-aminobutyric acid (GABA) indicated that TRPV1 was distributed on both glutamatergic and GABAergic neurons. Moreover, nerve growth factor, a sensitizing factor for TRPV1, was showed a higher expression pattern in MTLE patients. Taken together, our findings suggest that the overexpression and distribution patterns of TRPV1 might be involved in the pathogenesis and epileptogenesis of human MTLE.

Hepatocyte-derived exosomes deliver H2AFJ to hepatic stellate cells and promote liver fibrosis via the MAPK/STMN1 axis activation

Summary: This study explores the effects of exosomal H2AFJ derived from hepatocytes on the activation of hepatic stellate cells (HSCs) and liver fibrosis. It was found that H2AFJ, released by hepatocytes through exosomes, promotes HSC migration and invasion by activating the MAPK/STMN1 signaling pathway, exacerbating liver fibrosis. Silencing H2AFJ attenuates liver fibrosis in mice, while restoration of STMN1 negates this effect. This study provides a potential therapeutic target for alleviating liver fibrosis.

Examining the roles of experiential avoidance and cognitive fusion on the effects from mindfulness to athlete burnout: A longitudinal study

Summary: This study examined the mediating roles of experiential avoidance and cognitive fusion in the relationship between mindfulness and athlete burnout. The results showed that mindfulness had direct effects on experiential avoidance and cognitive fusion, which in turn influenced athlete burnout. These findings have important implications for reducing athlete burnout.

Association of Lateralized Interictal Epileptiform Discharges with Postsurgical Seizure Outcomes in Patients with Tuberous Sclerosis Complex: A 10-Year Retrospective Study at a Single Center

Summary: This study retrospectively analyzed the clinical characteristics and postsurgical seizure outcomes of 82 TSC patients, exploring the relationship between interictal epileptiform discharges (IEDs) and postsurgical outcomes. Results showed a significant association between IEDs and long-term postsurgical outcomes in TSC patients, with patients exhibiting focal, lateralized, and generalized IEDs achieving seizure freedom in the 1-year follow-up. Interestingly, patients with lateralized IEDs showed long-term seizure freedom in the 5-year and 10-year follow-ups. According to the study, IEDs may guide clinical care for TSC patients with intractable epilepsy.

Individualized portal pressure gradient threshold based on liver function categories in preventing rebleeding after TIPS

Summary: This study aimed to find the optimal threshold of individualized portal pressure gradient (PPG) to balance rebleeding and overt hepatic encephalopathy. The study found that different PPG standards may be required for patients with different liver function categories. For patients in Child-Pugh class B, a PPG threshold <12 mmHg might be more suitable, while <14 mmHg might be optimal for patients in Child-Pugh class C.

Clinical outcomes of clip-assisted endoscopic cyanoacrylate injection versus conventional endoscopic cyanoacrylate injection in treating gastric varices with a gastrorenal shunt

The results showed that clip-ECI was more effective than con-ECI, requiring fewer treatment sessions and achieving a lower 6-month rebleeding rate.

Promoting influenza prevention for older adults using the Health Action Process Approach: A randomized controlled trial

Effects of Habit and Intention on Behavior: Meta-Analysis and Test of Key Moderators

Summary: In a meta-analysis, the researchers aimed to estimate the size and variability of habit-behavior and intention-behavior relations while investigating theory-consistent moderators. They found that habit and intention independently predicted behavior, with habit partially mediating past-future behavior relations. Larger habit-behavior relations were observed in studies targeting behaviors with high habit formation opportunity and lower complexity. The findings support and extend previous research on habit and its measures.

Hippocampal ripples correlate with memory performance in humans

Summary: Memory performance evaluation has traditionally relied on behavioral tests, but the neural correlates, particularly in humans, have been unclear. In this study, we used intracranial electroencephalography in patients with refractory epilepsy to investigate the neural correlates of memory performance. We found that hippocampal ripples increased during visual stimulation or before verbal recall, and this increase was positively correlated with memory performance. On the other hand, memory performance was negatively correlated with hippocampal interictal epileptic discharges or epileptic ripples during the memory task. However, these correlations were not present during quiet wakefulness. Thus, our findings reveal the neural correlates of memory performance beyond traditional behavioral tests.

Integrating perceived physical environments and the theory of planned behaviors when explaining adherence to 24-hour movement guidelines in Chinese adolescents

Summary: This study aimed to investigate the associations between perceived physical environments, constructs of the theory of planned behavior (TPB), habit strength, and adherence to 24-h movement guidelines in Chinese adolescents. The findings showed that attitude, subjective norm, and perceived behavioral control had direct effects on intention, and intention had a direct effect on the number of guidelines being met. Habit strength was a significant predictor of adherence to the guidelines.
