Mechanisms of adrenal gland growth: signal integration by extracellular signal regulated kinases1/2

The adrenal gland influences a multitude of processes during stress response, but also potently affects the immune system, glucose metabolism, electrolyte or water homeostasis, and cardiovascular functions. According to the present understanding, the adrenal cortex is tightly controlled by the hypothalamic-pituitary-adrenal axis. This axis involves hypothalamic CRH and pituitary ACTH which determine processes of adrenocortical growth and function. However, control of the adrenal gland comprises a plethora of additional endogenous or exogenous factors. Among those are diverse hormones, psychosocial parameters, physiological stress, secondary plant products, or even environmental pollutants. In the present review, we summarize the current view of endocrine growth control in the adrenal gland. We then discuss intracellular mechanisms of adrenal growth control and focus on extracellular signal regulated kinases 1/2 (ERK1/2), which have been demonstrated to be controlled by not only ACTH or angiotensin 11, but also by a large number of additional effectors. On the basis of these multiple exogenous or endogenous factors which impact on the adrenal gland through ERK-1/2 activity, we speculate on a mechanism by which ERK-1/2 act as a central integrative growth regulatory elements in the adrenal gland. Summary: This study investigated the impact of maternal supply of essential fatty acids and conjugated linoleic acid on glucose metabolism, endocrine regulation, energy metabolism, and intestinal development in neonatal calves. The results showed that a combination of EFA and CLA may improve the glucose metabolism and insulin response in calves.

Using IGFBP2 to enhance the sensitivity of IGF-dependent bioactivity, phosphorylation of AKT-Ser473 was assessed as a readout of pathway activity.

Summary: The study investigated the effects of training and genetic predisposition on mRNA expression in the pituitary gland of a marathon mouse model, revealing a strong impact of genetic selection for enhanced running performance on gene expression in the pituitary gland. Training induced differential expression of genes in both mouse lines, but DUhTP mice showed resistance to the positive effects of training on protein and energy metabolism in the pituitary gland.

Summary: The study investigated the regulation of the GH/IGF system in physical performance using a mouse model selected for high running performance. Results showed coordinated downregulation of the somatotropic pathway and a lack of mTORC1 and mTORC2 activation in marathon mice in response to exercise. Additionally, the study found enhanced expression of certain genes related to endurance exercise in the muscle of the selected marathon mouse model.

Summary: This study investigates the effect of long-term high-intensity exercise on Satellite cells (SATC) in mice. The results show that mice trained in high-intensity running have higher SATC activity and proliferation rate, indicating increased sensitivity to external stimuli and an easier transition into differentiation. Additionally, these trained mice produce more large myotubes. The findings suggest that long-term high-intensity exercise can promote muscle growth by altering the characteristics of SATC.

Summary: In addition to being energy sources, monosaccharides are used to construct complex oligo- and polysaccharides, which have important roles as functional biomolecules. These bioactive sugars are key components of milk and have positive impacts on intestinal development, gut microbiome, immune system, and learning and memory ability of offspring. Additionally, milk oligo- and polysaccharides have anti-adhesive properties against pathogens, making them important for mammary gland and offspring health.
