Summary: The study demonstrates that the activation of CaMKII delta through Met281/282 oxidation is not necessary for the recovery of Ca2+ transients during acidosis or for the occurrence of postacidic cellular arrhythmias. Despite an usually prooxidant increase in cytosolic Na+, acidosis reduces the cytosolic glutathione redox state within cardiac myocytes, suggesting that oxidation of cytosolic proteins is less likely to occur during acidosis.

Inherited arrhythmia disorders caused by mutations in Ca2+-handling proteins are also discussed.

Summary: The contraction of heart cells is regulated by the signaling interaction between LCCs and RyRs, which is influenced by the nanodistance between JPH2 in the SR and CAV3 in the TT. Our study revealed that SRF and myocardin concurrently regulate the expression of JPH2 and CAV3, leading to enhanced efficiency of LCC-RyR signaling and compensation for LCC down-regulation during hibernation, maintaining heart power and avoiding calcium overload.

Summary: This study developed a tamoxifen-inducible NCX1 knockout mouse model, finding that NCX1 KO mice adapt to acute genetic ablation by reducing Ca2+ influx, increasing alternative Ca2+ efflux pathways, and increasing excitation-contraction coupling gain to maintain contractility at the cost of mild hypertrophy and fibrosis, with decreased survival. However, KO myocytes are protected against spontaneous action potentials and ischemia/reperfusion injury.

The sites of palmitoylation are found in the N terminus and the linker between domains I and II of the channel. Unpalmitoylated α1C subunit leads to reduced voltage sensitivity of Ca2+ channels.

Summary: This study suggests that transient elevation of blood glucose for 1 hour in non-diabetic hearts under higher Ca2+-load reduces contractile properties probably by activating CaMKII through O-GlcNAcylation. Therefore, in patients with severe disease, such as acute myocardial infarction or acute heart failure, transient elevation of blood glucose could worsen cardiac function and affect mortality without known diabetes.

Summary: This study investigated the relationship between chronic total occlusion (CTO) in a non-infarct-related artery (NIRA) and adverse outcomes in ST-segment-elevation myocardial infarction (STEMI). The presence of CTO posed a threat to coronary blood supply, and the origin of intercoronary collaterals was associated with patient survival rate. The results showed that CTO collateralization from the IRA distal to the acute occlusion was associated with a better prognosis.

Summary: The study aimed to evaluate the association between common diseases and nocturnal hypoxemic burden in patients undergoing coronary artery bypass grafting surgery. The results showed that sleep-disordered breathing, obesity, and mild-to-moderate chronic obstructive pulmonary disease were significant predictors of increased nocturnal hypoxemic burden.

Summary: mTOR, a mechanistic target of rapamycin, is an evolutionarily conserved pathway that plays a fundamental role in nutrient sensing, growth, metabolism, lifespan, and aging. Recent studies have highlighted the regulatory role of mTOR in innate immune responses and its involvement in the pathogenesis of cardiovascular diseases, especially in acute inflammation and atherosclerotic cardiovascular disease. This review also discusses mTOR's role in trained immunity, immune senescence, and clonal hematopoiesis, as well as its architecture and regulatory complexes.
