Summary: Novel (E)-2-benzylidene-N-(3-cyano-4,5,6,7-tetrahydrobenzo[b]thiophen-2-yl) hydrazine-1-carboxamide derivatives were designed and synthesized in this study, showing promising inhibitory activity against alpha-glucosidase. The most potent compound 7 exhibited non-competitive inhibition towards alpha-glucosidase and demonstrated in vivo hypoglycemic activity in sucrose-treated rats.

Summary: A new potent analgesic, (3R,4S)-9d, with low ED50 values in hot plate and antiwrithing models has been identified through phenotypic screening. Mechanistic studies revealed that its analgesic effect is mediated via the active metabolite (3R,4S)-10a, with proposed activation of the mu opioid receptor (MOR) mechanism using molecular dynamics (MD) simulation.

Summary: In this study, a retro-Claisen reaction was performed to synthesize compound 5, and a plausible mechanism was proposed. The crystal structure of the compound was confirmed using single-crystal X-ray diffraction analysis, revealing a layered structure. Experimental results and DFT and Hirshfeld surface analysis supported the synthesized compound. Further investigation suggested that compound 5 may serve as a carbonic anhydrase I inhibitor.

Summary: A novel series of 4,6,7-trisubstituted quinoline analogues containing thiazolidinones were designed and synthesized in this study. Compound 15i was identified as a potent multi-kinase inhibitor with significant antitumor activities against HT-29 cells. It induced apoptosis and cell cycle arrest in a dose- and time-dependent manner, while showing low toxicity to normal cells. Further structural modifications may lead to the development of more potent kinase inhibitors for cancer treatment.

Summary: A series of novel benzamide derivatives were designed and synthesized from the pyridazinone scaffold. Among them, (S)-17b showed potent inhibitory activity against human class I HDAC isoforms and human myelodysplastic syndrome (SKM-1) cell line in vitro. Furthermore, (S)-17b demonstrated excellent in vivo antitumor activity in SKM-1 xenograft models, and showed better efficacy on mouse models with intact immune system compared to those with thymus deficiencies. This novel compound (S)-17b may serve as a promising drug candidate for further investigation.
