Summary: Proteolysis-targeting chimera (PROTAC) is a promising technology in targeted protein degradation and drug discovery. This study designed the first fluorescent and theranostic PROTACs for real-time imaging of the degradation of nicotinamide phosphoribosyltransferase (NAMPT) in living cells. The results showed efficient degradation of NAMPT and significant reduction of nicotinamide adenine dinucleotide (NAD(+)), leading to potent antitumor activities in vitro and in vivo. This proof-of-concept study provides a new strategy for real-time visualization of protein degradation and improvement of diagnosis and therapeutic efficacy of PROTACs.

The results showed that patients with ACC had significantly higher visfatin serum concentrations compared to patients with benign adrenocortical tumors, and visfatin levels were associated with clinical stage and metastases in ACC patients. Serum visfatin/NAMPT could potentially serve as a biomarker for distinguishing ACC, but further research is needed to confirm its accuracy and reliability.

Summary: This study aimed to validate a U.S.-validated indicator for spontaneous preterm birth in populations from Bangladesh, Pakistan, and Tanzania, and to identify additional biomarkers that enhance its performance. The study found that the initial IBP4/SHBG biomarker predicted preterm birth, and the addition of endoglin, prolactin, and tetranectin improved the prediction. The findings suggest potential broader applications for the U.S.-developed biomarker in diverse non-U.S. populations.
