Summary: Several thiouracil amide compounds were synthesized and screened for their anticancer properties against human breast cancer cells. Some compounds exhibited moderate to significant efficacy, inhibited the catalytic activity of PARP1, enhanced DNA repair and apoptosis, and interacted with PARP1. Therefore, thiouracil amides have the potential to be developed as new drug seeds for PARP inhibitors in oncology.

Summary: This study describes a metal- and additive-free visible light-induced [4+2] annulation reaction to synthesize tetralin-1-carboxamide derivatives from acrylamides and 2-benzyl-2-bromocarbonyls. The developed method is convenient, efficient, and environmentally friendly, and can also be used for the synthesis of bridged polycyclic imides with excellent cis-diastereoselectivities.

Summary: The WNT/beta-catenin pathway plays a crucial role in regulating a wide range of cellular functions, and its dysregulation is closely associated with the development of cancer. In this study, the interaction between the protein Dishevelled 1 (DVL1), a key player in this pathway, and its receptor Frizzled was investigated. Through computational studies, a compound called RS4690 (1) was discovered to selectively inhibit the binding of DVL1. After separating the racemic mixture, the enantiomer (S)-1 showed better inhibition of DVL1 and the growth of HCT116 cells, indicating its potential as a new therapeutic agent against WNT-dependent colon cancer.
