Summary: alpha-Deuterated amino acids are valuable for developing deuterated drugs and studying biological systems. This study demonstrates that a PLP-dependent Mannich cyclase, LolT, can catalyze the deuteration of a diverse range of L-amino acids with high site- and stereoselectivity. The researchers also discovered that LolT exhibits beta-elimination activity with certain amino acids, confirming previous hypotheses about its evolution. The versatility and selectivity of LolT make it a powerful biocatalyst for preparing alpha-deuterated L-amino acids.

Summary: This study demonstrated the protective effect of membrane-free stem cell extract (MFSCE) on cognitive impairment in Alzheimer's disease (AD) mice injected with A beta(25)(-)(35). The combination treatment of MFSCE and pyridoxal 5'-phosphate (PLP) showed greater suppression of apoptotic signaling and amyloidogenic-pathway-related proteins, leading to improved cognitive function in AD. The antioxidant effects of MFSCE were associated with inhibition of lipid peroxidation and nitric oxide overproduction, highlighting the potential therapeutic implications for AD patients.

ObiH, an l-threonine transaldolase, demonstrated the ability to selectively synthesize diverse non-standard amino acids on a scalable whole cell platform. Results showed high initial selectivity of the catalyst and a decrease in diastereomeric ratio of products at high conversion.

Summary: In silico modeling was used to identify a selective inhibitor for ALDH1A3, a potential drug target highly expressed in mesenchymal glioma stem cells. The identified inhibitor, MCI-INI-3, showed efficient inhibition of retinoic acid biosynthesis comparable to ALDH1A3 depletion. Further research is needed to understand the role of ALDH1A3 and retinoic acid biosynthesis in glioma stem cell growth and differentiation.

Summary: Aldehyde dehydrogenase 1A3 (ALDH1A3) has been linked to poor prognosis in malignant pleural mesothelioma (MPM). Inhibition of ALDH1A3 using NR6 resulted in the accumulation of toxic aldehydes, DNA damage, CDKN2A expression, and cell growth arrest. NR6 treatment also affected the tumor microenvironment by altering IL6, CXCL8, and IL-8 levels, and shifting cell fate from senescence to apoptosis. Understanding the role of ALDH1A3 in MPM cells and the tumor microenvironment could lead to potential new treatments for this cancer.
