Summary: A metal, solvent and oxidant free method has been developed for the synthesis of quinazolin-4(3H)-ones under microwave irradiation. Using the same catalyst, 2,3-dihydroquinazolin-4(1H)-ones were also synthesized in water as a green and sustainable solvent. These protocols offer a simplistic and rapid approach for assembling diverse structural quinazolinones.

Summary: A nickel-catalyzed cross-electrophile coupling reaction between 1,2,3-benzotriazin-4(3H)-ones and aryl bromides has been developed, leading to a diverse array of ortho-arylated benzamide derivatives. The reaction exhibits good functional group tolerance and utilizes the ring opening of benzotriazinones followed by denitrogenation to obtain various benzamide derivatives (29 examples, 42-93% yield). The scalability of this transformation has been demonstrated.

Summary: Four novel 3-Aryl-1-(pyridin-4-yl)benzo[4,5]imidazo[1,2-d][1,2,4]-triazin-4(3H)-ones derivatives were designed, synthesized, and evaluated for their anticancer activity. The structures of the compounds were characterized and their binding to EGFR was determined. One compound, C3, exhibited the highest cytotoxicity against breast cancer cells and also inhibited cell migration. Western blotting results indicated that C3 treatment inhibited EGFR signaling, induced cell cycle arrest and apoptosis. This study provides a promising starting point for the development of new compounds targeting cancer cells.

Summary: Antifolates are a class of drugs used for antibacterial, antiparasitic, and anticancer purposes. 2-substituted-mercapto-quinazolin-4(3H)-one analogues have been studied as DHFR inhibitors, with the development of compounds with significant biological activity based on a structural model. The compounds 18, 20, and 21, formulated using this model, have shown 4-8 times greater activity than the reference drug methotrexate (MTX, 1).

Summary: Highly functionalized quinazolin-4(3H)-ones were synthesized from reactions of N-aryl-2-aminobenzamides with trialkylamines under photocatalytic conditions using eosin Y as a catalyst. The reaction was mild and carried out in aqueous acetonitrile, showing a broad substrate scope. Mechanistic studies revealed the electron-donor nature of the intermediate 2,3-dihydroquinazolin-4(1H)-one (3') in the photocatalytic cycle, leading to the formation of 2,3-disubstituted quinazolin-4(3H)-ones (3). This methodology was also successfully extended to the synthesis of benzo [4,5] imidazole[1,2-c] quinazolines and 3 aa on a large scale, as well as the CNS depressant drugs methaqualone (3 la) and mecloqualone (3 pa).

Summary: A green, simple and efficient method for the synthesis of quinazolin-4(3H)-ones has been developed via visible light-induced condensation cyclization. The method utilizes fluorescein as a photocatalyst and TBHP as a reaction promoter without the need for a metal catalyst. The reaction demonstrates high substrate tolerance and yields a variety of desired products in good to excellent yields. This method provides a straightforward strategy for the synthesis of quinazolin-4(3H)-ones.

The influence of different donor fragments on fluorescence quantum yields and emission intensity is demonstrated. Some compounds show enhanced fluorescence intensity in response to the addition of water in solution.

Summary: In this study, we achieved a C3-selenylation of pyrido[1,2-a]pyrimidin-4-ones using an electrochemically driven external oxidant-free strategy. Various structurally diverse seleno-substituted N-heterocycles were obtained in moderate to excellent yields. A plausible mechanism for this selenylation was proposed based on radical trapping experiments, GC-MS analysis, and cyclic voltammetry study.

This protocol is characterized by high efficiency, excellent migrating selectivity, broad substrate scope, and convenient one-pot procedure, making it an effective tool for piperidine derivative synthesis. The migration-annulation reaction of α-imino carbene presents a powerful strategy for heterocycle construction.

Summary: Three sets of new quinazolinones were synthesized and evaluated for their cytotoxicity against cancer cell lines. Compounds 4b, 4d, 6f, 6g, 8c, 8f, and 8g showed promising activity as VEGFR-2 inhibitors. Among them, compound 8f exhibited potent inhibitory effects on multiple signaling pathways and demonstrated low toxicity against normal cells.

The catalyst demonstrated excellent catalytic efficiency, magnetic recoverability, and reusability for multiple cycles, making it a promising candidate for sustainable and efficient catalysis in organic synthesis.
