Streptococcus urinalis was isolated from a blood culture of a 60-year-old man with a history of urethral stricture. This species has been recently described as a new member of the pyogenic subgroup of streptococci that cause urinary tract infections.

Case report: One human Streptococcus suis occurred in Shandong Province, China

Summary: A simple viscoelastic-based microfluidic platform is presented for label-free isolation of small extracellular vesicles (sEVs) from human blood, with high purity, recovery rates, and increased yield compared to conventional methods. The clinical utility of the approach is demonstrated by isolating sEVs from blood samples of cancer patients and healthy donors, showing elevated sEV concentrations in the blood of cancer patients.

Isolation methods determine human neutrophil responses after stimulation

Summary: This study assessed the impact of five commonly used methods for neutrophil isolation on neutrophil yield, purity, activation status, and responsiveness. The results showed that neutrophils isolated by negative immunomagnetic selection and density gradient methods, without red blood cell lysis, closely resembled untouched neutrophils in whole blood. These neutrophils were less activated and more responsive to milder stimuli compared to those obtained using density gradients requiring red blood cell lysis. The study highlights the importance of selecting the appropriate method for studying neutrophils and standardizing isolation protocols.

Summary: The neurovascular unit (NVU) is crucial for the proper functioning of the central nervous system and involves endothelial cells, pericytes, astrocytes, microglia, and neurons. Dysfunction of the blood-brain barrier (BBB), regulated by the NVU, is seen in various neurological diseases and is a major therapeutic target. In this study, the EPAM-ia method is introduced as a protocol for simultaneous isolation and analysis of NVU cells using flow cytometry. This method involves the differential processing and combination of NVU cell types followed by immunostaining and fluorescence-activated cell sorting. The EPAM-ia method has applications in transcriptomics, proteomics, and cell culture and can be employed for analyzing various cell types in neurological diseases associated with BBB dysfunction.

A Simplified and Effective Approach for the Isolation of Small Pluripotent Stem Cells Derived from Human Peripheral Blood

Summary: Pluripotent stem cells play a vital role in regenerative medicine. However, the limited availability and ethical concerns associated with embryonic pluripotent stem cells have led to the discovery of very small embryonic-like (VSEL) stem cells. In this study, a simplified and effective method for isolating small pluripotent stem cells from human peripheral blood is presented. The isolated small blood stem cells (SBSC) population expresses pluripotency markers and also exhibits characteristics of hematopoietic and mesenchymal stem cells. Proteomic profiling reveals the presence of various stem cell markers and transcription regulatory complex factors in SBSCs. This novel isolation process yields a abundant population of small-sized cells with pluripotent characteristics from human peripheral blood.

Large inter-individual variability of cellular and humoral immunological responses to mRNA-1273 (Moderna) vaccination against SARS-CoV-2 in health care workers

Summary: This study conducted a longitudinal immunological analysis of healthcare workers vaccinated with Moderna mRNA-1273 vaccine. The results showed significant inter-individual differences in cellular and humoral immune responses. The study suggests the need for serological assays to verify the success of the vaccine and demonstrates the correlation between antibody kinetics and clinical effectiveness, explaining the mechanism of vaccine efficacy in COVID-naive individuals mediated by low-avidity antibodies.

Summary: The rapid pace of name changes of medically important fungi creates challenges for clinical laboratories and clinicians. Suggestions are made to reduce the number of name changes and provide diagnostic markers. It is recommended to maintain stability at the genus level and report both overarching species and molecular siblings. An open access online database of all medically important fungi names is essential, and new name changes should be reviewed routinely by a standing committee.
