Summary: The study found that early CRPS IgM antibodies have pronociceptive effects in fracture mice, potentially by activating C5a complement signaling pathway leading to increased proinflammatory cytokine expression in the skin and spinal cord, contributing to nociceptive sensitization in the injured limb.

Summary: Bisphosphonate-related osteonecrosis of the jaw (BRONJ) is a serious side effect of systematic administration of bisphosphonates (BPs). Sensory denervation facilitates the occurrence of BRONJ, but therapeutic use of CGRP can effectively prevent the progression of BRONJ.

Summary: Postoperative delirium is common in elderly patients with hip fracture, and pain is a major risk factor for delirium. Regional nerve blocks (RNBs) are effective in controlling pain and reducing delirium incidence. A study involving 252 patients showed that RNB was associated with lower delirium rates and better pain control in elderly patients undergoing hip surgery.

Summary: In this study, it was found that increased expression of SP and CGRP in biopsy samples of lateral epicondylitis patients was associated with tendon degeneration severity. The levels of SP and CGRP expression were significantly correlated with Movin score, with higher levels associated with more severe degeneration. Additionally, elevated SP or CGRP expression levels were also linked with higher preoperative VAS scores.

Summary: In tibial plateau fracture surgery, regional anesthesia (RA) is associated with decreased inpatient opioid consumption and reduced outpatient opioid demand. The use of RA could be a viable option to consider.

Summary: Alpha CGRP and substance P play important roles in bone homeostasis, and their ratio is crucial for driving aging and healing scenarios. By increasing the alpha CGRP/SP ratio, better callus formation and faster callus remodeling can be achieved during fracture healing.

Summary: Periradicular tissues contain trigeminal nociceptors that play a role in both relaying pain signals and modulating immune responses in apical periodontitis. Studies have investigated the neuroanatomy and biomolecules profiles to understand the contributions and mechanisms of these nociceptors in inflammation and tissue regeneration. This narrative review aims to address the complex interactions between trigeminal nociceptors and macrophages in the clinical manifestations of apical periodontitis.

Summary: Complex regional pain syndrome (CRPS) is a rare and debilitating chronic pain disorder characterized by persistent pain that is disproportionate to any preceding injury. It can significantly impact a person's quality of life, resulting in disability and psychological distress. Despite being recognized for over a century, finding effective treatment for CRPS remains challenging.

Summary: Cizolirtine, a substance used for pain and urinary incontinence, showed an increased incidence of tumors in mice and rats. In mice, it was associated with an increase in skin fibrosarcomas and sarcomas, while in rats, it was associated with liver adenomas and carcinomas, possibly due to increased liver metabolism and related changes. However, the relevance of these findings to humans remains uncertain.

Summary: Cizolirtine treatment in rats showed adverse effects such as decreased bodyweight gain and increased plasma concentrations. High doses led to liver and ovarian issues in some animals. The dosages of 20 and 60 mg/kg/day were considered as the No-Observed-Adverse-Effect Levels for males and females respectively.

Summary: This study found a close association between the recurrence of pterygium and increased levels of Substance P and Vasoactive Intestinal Peptide (VIP) in TRPV1-positive nerve fibers. Changes in TRPV1 expression were also observed in recurrent cases. These findings suggest a potential role of TRPV1 in pterygium recurrence and indicate that inhibiting TRPV1 activity may help prevent pterygium recurrence.

Summary: This study investigated the detailed morphological characteristics of mandibular canal branches (MCBs) using CBCT and observing the distribution of CGRP around the MCB. It found correlations between the structure of MCBs and CGRP distribution, with most MCBs following a clockwise distribution pattern and positive CGRP reactions observed in the no-canal branch group. These findings provide valuable insights for MCB dynamics and clinical dental treatment.

Summary: The study evaluated the safety of intravesical application of resiniferatoxin (RTX) in healthy cats and its effects on calcitonin gene-related peptide (CGRP) and substance P (SP) produced by C-fibers. The results showed that a dose of 25 μg (1 μg/mL) of RTX reduced the innervation of nerve fibers in the bladder and induced moderate changes in the bladder tissue.
