Role of CRD-BP in the Growth of Human Basal Cell Carcinoma Cells

Although the number of new cases of basal cell carcinoma (BCC) has increased rapidly in the last few decades, the molecular basis of its pathogenesis is not completely understood. Activation of the Hedgehog (Hh) signaling pathway has been shown to be a key factor driving the development of BCC. The Wnt/beta-catenin signaling pathway was also shown to be activated in BCCs and to perhaps modulate the activity of the Hh pathway. We have previously identified a mechanism by which Wnt signaling regulates the transcriptional outcome of the Hh signaling pathway. We demonstrated that coding region determinant-binding protein (CRD-BP), a direct target of the Wnt/beta-catenin signaling, binds to GM mRNA, stabilizes it, and consequently upregulates its levels (mRNA and protein) and activities. We hypothesized that Wnt-induced and CRD-BP-dependent regulation of GEM expression and activities is important for the development of BCC. In this study, we show that CRD-BP is overexpressed in BCC and that its expression positively correlates with the activation of both Wnt and Hh signaling pathways. We also describe the generation and characterization of a human BCC cell line. This cell line was utilized to demonstrate the importance of CRD-BP-dependent regulation of GM expression and activities in the development of BCC.

Summary: IKAROS functions as a transcriptional repressor of MTOR gene in treating HR B-ALL, which can be enhanced by the CK2 inhibitor CX-4945. The combining of CX-4945 with rapamycin shows novel synergistic therapeutic effects in reducing MTOR expression and treating Hispanic/Latino children with HR B-ALL. Dual targeting of oncogene transcription along with oncoprotein inhibition could provide a precision medicine approach for hematological malignancies.

Small extracellular vesicles induce resistance to anti-GD2 immunotherapy unveiling tipifarnib as an adjunct to neuroblastoma immunotherapy

Summary: This study investigated the role of neuroblastoma-derived small extracellular vesicles (sEVs) in promoting resistance to anti-GD2 monoclonal antibody therapy and the potential of tipifarnib in sensitizing tumors to the antibody treatment. The results showed that sEVs attenuated the efficacy of the antibody and modulated the tumor microenvironment and immune cell infiltration. Additionally, tipifarnib enhanced the antibody's effectiveness against tumor growth and prevented the immunosuppressive effects of sEVs.

Summary: Iron is vital for brain development and function. This study investigated the role of exosomes in transporting iron across the blood-brain barrier endothelial cells (BBBECs). The results showed that iron activity in the exosome fraction was higher than in the supernatant fraction. The release of exosomes containing iron was regulated by the iron concentration in the endothelial cells and was independent of hepcidin regulation.

IGF2BP1 regulates the cargo of extracellular vesicles and promotes neuroblastoma metastasis

Summary: Neuroblastoma (NB) is a highly metastatic cancer that causes a high number of cancer-related deaths in pediatric patients. Up to 50% of NB cases show a partial chromosomal gain at locus 17q21-ter, which is associated with poor survival. This study investigates the role of the proto-oncogene IGF2BP1, located at 17q locus, in promoting NB metastasis and its regulation of small extracellular vesicles (EVs). The study identifies SEMA3A and SHMT2 as novel targets of IGF2BP1 and reveals the mechanisms by which IGF2BP1 contributes to NB metastasis through regulating protein levels in NB-EVs. The findings suggest the clinical significance of IGF2BP1-SEMA3A/SHMT2 axis in NB metastasis.

Apo- and holo-transferrin differentially interact with hephaestin and ferroportin in a novel mechanism of cellular iron release regulation

Summary: Using cell culture techniques, this study investigates the mechanism by which apo- and holo-Tf influence iron release from endothelial cells. It is found that holo-Tf induces ferroportin internalization, while apo-Tf directly interacts with hephaestin. Only pathophysiological levels of hepcidin disrupt the interaction between holo-Tf and ferroportin. These findings provide a molecular mechanism for the regulation of iron release.
