Summary: Epiregulin-EGFR signaling between dendritic cells and fibroblasts plays a critical role in maintaining elevated ECM production and accumulation in fibrotic tissue. Epiregulin is induced by type I interferon and activates EGFR on fibroblasts through NOTCH signaling, driving a positive feedback loop. Epiregulin is essential for persistence of fibrosis in skin and lung tissues, and therapeutic administration of epiregulin antibody can reverse fibrosis.
