Summary: Cell-fate decisions rely on the regulation of multiple signaling molecules and transcription factors. IP3R3, a tetrameric channel, releases Ca2+ in response to extracellular stimuli. Its gating is regulated by ligands and interacting proteins. Understanding the structure and regulation of IP3R3 provides insights into its biological functions and pathogenic mechanisms.

Summary: The synergistic role of Foxp1/4 in maintaining the quiescence of HFSCs and the stem cell niche is revealed. The double deficiency of Foxp1/4 leads to premature activation of HFSCs and hair loss, accompanied by the destruction of the stem cell niche.

Summary: A peptide called ICT60 was designed to inhibit the aggregation and cytotoxicity of the huntingtin protein (Htt) in Huntington's disease (HD) by interacting with IP3 receptor 1 (IP3R1). The results provide a molecular basis for developing therapeutics to treat HD.

Summary: This study aimed to explore the impact of the Hippo/YAP signaling pathway on cochlear hair cell injury by regulating ferroptosis. The results showed that cisplatin inhibited the viability of HEI-OC1 cells by increasing free Fe2+ levels and decreasing FTL levels. LAT1-IN-1 promoted cell viability by suppressing oxidative stress and ferroptosis, while verteporfin had the opposite effect. YAP1 transcriptionally regulated the expression of FTL and TFRC. Inhibition of FTL or TFRC affected cell viability and ferroptosis. In conclusion, YAP1 ameliorated cochlear hair cell injury by upregulating FTL and TFRC to suppress ferroptosis.

Summary: Two new human MRAP2 splice variants were identified in this study, and the effects of MRAP1 and MRAP2 on MC3R and MC4R were investigated. The results suggest that MRAPs play important roles in the signaling and regulation of neural MC3R and MC4R.

Summary: This study investigates a novel imidazobenzodiazepine called PI320 and its potential as an asthma drug. The results show that PI320 can alleviate bronchoconstriction and relax peripheral airways in asthma models. The mechanism of action appears to involve inhibiting intracellular calcium mobilization in airway smooth muscle.

Summary: This study found that CD200 protein in basal cell carcinoma may lead to the near absence of NK cells in the tumor microenvironment through ectodomain shedding. CD200 suppresses MAPK signaling in NK cells, blocking indirect and direct cell killing. Blocking CD200 inhibition can restore NK cell killing ability.

Summary: In fruit fly larvae, the IP3/Ca2+ signals in specific interneurons drive the expression of Set2 gene, essential for pupariation in the absence of dietary protein. This signal-controlled expression of Set2 upregulates key Ca2+-signaling genes through a positive-feedback loop, regulating gene expression for the transition from larval to pupal stages.

Summary: Early research suggested a simple relationship between phosphate exposure, increased serum FGF23 concentrations, and clinical endpoints. However, subsequent studies have revealed a more complex interplay between factors in bone. In addition to physiological regulators, various disease states also influence FGF23 levels, increasing the burden of disease. While some factors may confound the association between FGF23 and clinical endpoints, others are in the same causal pathway and hold promise for future therapeutic strategies.
