These esterified vitamin D3 make up a large fraction of tissue vitamin D3 in mice, but are hydrolyzed before being transported into the circulation.

By encapsulating into PLGA nanoparticles, 7-DHC shows efficient delivery and significantly enhances the efficacy of radiotherapy, leading to tumor eradication without side effects.

Summary: This study achieved highly efficient 7-DHC synthesis by remodeling the complicated sterol synthesis modules. By overexpressing multiple genes and a transcription factor mutant, as well as applying CRISPR-mediated activation and repression systems, the titer of 7-DHC was successfully increased. Furthermore, by compartmentalizing enzymes and enlarging the endoplasmic reticulum lumen, the titer of 7-DHC was further enhanced, reaching the highest reported level so far.

Summary: Anemarrhena asphodeloides Bunge, an important Traditional Chinese Medicine herb containing high levels of total steroidal saponins, primarily accumulates timosaponins in the rhizomes. Analysis of candidate genes involved in the timosaponin biosynthetic pathway revealed their high expression in the rhizomes, consistent with metabolic analysis results. Cloning and expression of two 7-dehydrocholesterol reductase genes in yeast supported their involvement in timosaponin biosynthesis. Phylogenetic analysis identified Aa7DR1 as a monocotyledonous Delta(7) reductase in phytosterol biosynthesis. This study expands our understanding of timosaponin biosynthesis.

Summary: This study utilized marker-independent long-term 3D live imaging and single-cell transcriptomics to investigate the development of epithelial lineage in mouse hair follicles. The findings revealed that precursors of different epithelial lineages align in a 2D concentric manner and extend to form longitudinally aligned, 3D cylindrical compartments. The fate of placode cells is determined by cell position rather than the orientation of cell division.

Summary: This study improved the biosynthetic and secretory productivity of 7-Dehydrocholesterol (7-DHC) in Saccharomyces cerevisiae by reinforcing and reengineering the ergosterol pathway genes, introducing a biphasic fermentation system, and overexpressing lipid transporters. The selective secretion of different sterol intermediates was characterized, and the cooperation between Pr-1 and St1 in sterol transport was confirmed. The improved cell biosynthetic capacity offers a novel modification approach for 7-DHC de novo biosynthesis.
