Immunogenicity and protective efficacy of a major White Spot Syndrome Virus (WSSV) envelope protein VP24 expressed in Escherichia coli against WSSV

The study reports cloning, expression and characterization of immunogenic activity of VP24, a major envelope protein of White Spot Syndrome Virus (WSSV). His-tagged VP24 was expressed as truncated protein and purified from inclusion bodies by metal affinity chromatography under denaturing conditions. The ability to confer protection from WSSV by oral administration of recombinant viral protein (rVP24) was examined in black tiger shrimp Penaeus monodon (P. monodon) juveniles (advanced post larvae). Animals were fed with rVP24 for 10 days, orally challenged with WSSV and assayed for expression of viral genes and shrimp immune genes on the 2nd, 5th and 8th days of challenge. The survival of juvenile shrimps in the vaccinated and challenged group was significantly higher compared to the unvaccinated and challenged group with lesser viral gene expression (DNA polymerase, latency 1 and vp28). Analysis of immune gene expression showed upregulation of syntenin and down regulation of STAT, Rab 7 and caspase during the experimental period. Nitrification and denitrification in recirculating aquaculture systems: the processes and players

Summary: One of the major challenges in sustainable aquaculture production systems is the accumulation of nitrogenous waste. By establishing in situ nitrification and denitrification through biofilters/bioreactors activated with nitrifying/denitrifying bioaugmentors, total nitrogen removal can be achieved without external carbon supplements or additional anaerobic compartment. The structure and population dynamics of nitrifying/denitrifying consortia in recirculating aquaculture systems are influenced by environmental factors, which are crucial for the success of the processes.

A Novel Approach of Transducing Recombinant Baculovirus into Primary Lymphoid Cells of Penaeus monodon for Developing Continuous Cell Line

Summary: The development of shrimp cell lines has been attempted for over 30 years, with one of the main challenges being replicative senescence and inability to maintain telomere length in vitro. By inducing oncogenic transformation through a recombinant baculovirus containing H-ras downstream of the shrimp viral promoter IHHNV-P2, a fibroblast-like primary cell culture with confirmed H-ras and GFP expression was successfully developed. Transduction efficiency during early dividing stage of cells can be enhanced by adding 5 mM sodium butyrate to the culture medium.

Summary: This study demonstrates the probiotic effect of bio-augmented Bacillus tequilensis AP BFT3 on the production, immune response, and proteomic changes of Penaeus vannamei reared in a biofloc system. Adding the probiotic in the biofloc rearing significantly improves shrimp growth, survival, and water quality. The expression of immune-related genes is upregulated, and differential protein expression is observed. Adding B. tequilensis AP BFT3 as a probiotic in a biofloc system reduces the incidence of diseases during culture.

Summary: Infectious hypodermal hematopoietic necrosis virus (IHHNV/PstDVI) was successfully isolated and propagated in a hybrid shrimp-insect cell line PmLyO-Sf9. Cytopathic changes such as clustering, enlargement, syncytium formation, and cell death were observed in the infected cell line, indicating viral replication. The purified virus was found to be infectious both in vitro and in vivo, providing insights for studying the molecular mechanism of IHHNV infection and developing antiviral strategies in aquaculture.

Summary: The recognition and elimination of invading pathogens are essential for host survival. Macrophages, which function similarly to vertebrate macrophages, play a crucial role in host protection in all multicellular organisms. CD14 is a pattern responsible for bacterial recognition found on the surface of macrophages. These cells store antimicrobial molecules and display polarization states similar to mammalian macrophages. The present study reports the presence of various cell markers, including CD14, in the intestinal macrophages of catfish and bonytongue, providing insights into the in vivo function of macrophages in fish.

Summary: The study found that the fish meal-based diet showed the best growth performance, followed by the pre-extruded Nannochloropsis diet. Feeding of unprocessed Nannochloropsis and Tetraselmis resulted in reduced enterocyte vacuolization. There was a significant effect of processing on fillet fatty acid content, intestine and liver structure, and the expression of selected genes in the liver.
