Picolinate ruthenium(II)-arene complex with in vitro antiproliferative and antimetastatic properties: Comparison to a series of ruthenium(II)-arene complexes with similar structure

In our previous study, ruthenium(II)-p-cymene complexes of general formula [(eta(6)-p-cymene)Ru(L)Cl2], L: 3-acetylpyridine (1), 2-amino-5-chloropyridine (2); and [(eta(6)-p-cymene)Ru(HL)Cl], HL: 2,3-pyridinedicarboxylic acid (3), 2,4-pyridinedicarboxylic acid (4), revealed low antiproliferative activity, except complex [(eta(6)-p-cymene)RuCl(picolinic acid)]center dot H2O (5) which exhibited IC50 around 80 mu M. In this study we further investigated in vitro potential of antimetastatic action of ruthenium complexes on HeLa and two endothelial cell lines. Comparison of structure and activity of five complexes indicated heterogenic mode of activity, with regard to the potential of antimetastatic and antiproliferative effect. Replacement of substituted pyridine ligand with picolinic acid (complex 5) around Ru(II) center contributed to complex cytotoxicity and ruthenium DNA binding affinity. Analysis of ruthenium(II) accumulation in DNA and protein fractions of HeLa cells, using ICP-OES revealed significantly higher content of complex 5 in DNA fraction in comparison to the other tested compounds. It also altered cell cycle progression, affected expression of DNA repair enzymes ERCC1 and MSH2, and showed enhanced activity in combination with 3-aminobenzamide. Regardless of their effect on cell growth, Ru(II) complexes exerted antimetastatic effect on several tumor cell lines in vitro, achieved mostly by the effect on cell adhesion, migration and angiogenesis, while picolinate ruthenium(II)-arene additionally exerted inhibitory effect on extracellular matrix degradation. Drug combination study of novel oxorhenium(V) complexes

Summary: In this study, three Re(V) complexes were synthesized and characterized, and their cytotoxicity against breast adenocarcinoma and pancreatic adenocarcinoma cells was investigated. One of the complexes, C1, showed dose-dependent cytotoxic potential and enhanced antiproliferative action in combination with an inhibitor of efflux transporter P-glycoprotein. Depletion of glutathione further increased the activity of C1.

(Electro)chemical and antimicrobial characterization of novel Ru(II) bipyridine complexes with acetylpyridine analogs

Summary: Three ruthenium-bipyridine complexes carrying acetylpyridine ligand unit were synthesized and characterized in this study. Their electrochemical activity, DNA-binding activity, antimicrobial activity, in vitro toxicity, and radical scavenging capacity were evaluated. The compounds showed moderate antimicrobial activity, no toxicity, and no antioxidant activity.

Cytotoxicity profiling of choline chloride-based natural deep eutectic solvents

Summary: This study investigated the cytotoxic profile of twelve choline chloride-based deep eutectic solvents (NADES) on HT-29, Caco-2, MCF-7, and MRC-5 cell lines. The cytotoxic effect was found to depend on the structure of the hydrogen bond donor (HBD). Acidic systems with organic acids as HBDs showed the highest cytotoxic effects, especially on the HT-29 cell line. The cytotoxicity was influenced by the concentration of the NADES system in the cell medium and the chemical constitution of the systems.

The Role of Isoflavones in the Prevention of Breast Cancer and Prostate Cancer

Synthesis, chemical characterization, and antimicrobial potency of picolinate-based half-sandwich Ru(II) complexes

Summary: Eleven new and three reported half-sandwich Ru(II) arene complexes were synthesized and evaluated for their in vitro antibacterial and antifungal activity. The structures and purity of the compounds were confirmed using various spectroscopic and analytical techniques. The ligands exhibited better inhibitory effect against Gram-positive bacteria compared to the metal complexes, while all synthesized complexes showed similar activity against Candida albicans.

Exploring heterometallic bridged Pt(II)-Zn(II) complexes as potential antitumor agents

Among them, C2 exhibited highly selective cytotoxicity and induced cell cycle arrest and expression of apoptotic related genes.

Breakthrough Analysis of Chemical Composition and Applied Chemometrics of European Plum Cultivars Grown in Norway

Summary: The aim of this study was to identify the chemical parameters for distinguishing plum cultivars grown in different regions of Norway. Chemical analysis of 68 plum cultivars revealed variations in the contents of elements, sugars, organic acids, phenolic compounds, and antioxidant activity. These differences have implications for human health and future breeding efforts.

In vitro and in silico study of the biological activity of tetradentate Schiff base copper(II) complexes with ethylenediamine-bridge

Structure prediction of physiological bis(amino acidato)copper(II) species in aqueous solution: The copper(II) compounds with L-glutamine and L-histidine

Summary: This paper investigates the geometries, energy landscapes, and magnetic parameters of copper(II) amino acid compounds using density functional theory calculations. The results show that certain conformers with histaminate-like and glycine-like modes have low Gibbs free energies and high metal-binding affinities. The predicted conformations reproduce the experimental electron paramagnetic resonance parameters.

Structural motifs in the early metallation steps of Zn(II) and Cd(II) binding to apo-metallothionein 1a

Summary: Many proteins require a metal cofactor for their function, and these metals play a role in protein folding. The study found that the metallation rates of different metal binding pathways are influenced by pH, with the less cooperative pathway being more affected by low pH conditions. Specific mixtures of structures are formed in the initial steps of metallation, and unfolding the disordered apo-MT structure hinders the formation of these preliminary structures. The compact conformation of the native apo-MT allows for rapid formation of metal-thiolate structures with high affinity, providing protection from oxidation.
