Signaling pathways involved in LPS induced TNFalpha production in human adipocytes

Background: The development of obesity has been linked to an inflammatory process, and the role of adipose tissue in the secretion of pro-inflammatory molecules such as IL-6 or TNFalpha has now been largely confirmed. Although TNFalpha secretion by adipose cells is probably induced, most notably by TLR ligands, the activation and secretion pathways of this cytokine are not yet entirely understood. Moreover, given that macrophagic infiltration is a characteristic of obesity, it is difficult to clearly establish the level of involvement of the different cellular types present within the adipose tissue during inflammation. Methods: Primary cultures of human adipocytes and human peripheral blood mononuclear cells were used. Cells were treated with a pathogen-associated molecular pattern: LPS, with and without several kinase inhibitors. Western blot for p38 MAP Kinase was performed on cell lysates. TNFalpha mRNA was detected in cells by RT-PCR and TNFalpha protein was detected in supernatants by ELISA assays. Results: We show for the first time that the production of TNFalpha in mature human adipocytes is mainly dependent upon two pathways: NFkappaB and p38 MAP Kinase. Moreover, we demonstrate that the PI3Kinase pathway is clearly involved in the first step of the LPS-pathway. Lastly, we show that adipocytes are able to secrete a large amount of TNFalpha compared to macrophages. Conclusion: This study clearly demonstrates that the LPS induced activation pathway is an integral part of the inflammatory process linked to obesity, and that adipocytes are responsible for most of the secreted TNFalpha in inflamed adipose tissue, through TLR4 activation.

Summary: This study evaluated the antioxidant and cytoprotective effects of polyphenol-rich extracts from two medicinal plants, A. borbonica and D. apetalum, from Reunion Island. The results showed that the polyphenol extracts of these plants could limit LDL oxidation and intracellular oxidative stress, and protect the endothelium.

Summary: This study evaluated the effects of polyphenols extracted from Antirhea borbonica on obesity and diabetes-related cerebrovascular damages during stroke. The results showed that polyphenols reduced insulin resistance, hyperglycemia, and dyslipidemia caused by obesity, and attenuated brain infarct, hemorrhagic transformation, and blood-brain barrier disruption. Polyphenols also exhibited anti-inflammatory and antioxidant properties, and decreased MMP-2 activity in hyperglycemic condition. Furthermore, the protective effects of polyphenols were associated with their circulating metabolites.

Summary: This study aimed to improve the bioavailability of curcumin and evaluate the synergistic effect of curcumin-reconstituted high-density lipoprotein nanoparticles on MGO-induced endothelial dysfunction in diabetic patients. The results showed that Cur-rHDL nanoparticles can alleviate MGO-induced cytotoxicity and oxidative stress, protect endothelial cell barrier integrity, and enhance the bioavailability of curcumin.

Structure-Based Virtual Screening and Discovery of New Bi-functional DAPK1 Inhibitors

Summary: Recently, a new signaling complex involving DAPK1 and NR2B was discovered to play a role in neuronal death cascade, where NMDA receptors interact with DAPK1 through NR2B subunit after stroke injury, leading to excitotoxicity. Using the ZINC-12 database, potential inhibitors of DAPK1 were identified, particularly natural compounds showing strong binding affinity with both ATP-binding site residues and substrate-recognition motifs. These findings suggest that these ligands have the potential to be inhibitors of DAPK1.
