Summary: Typhoid Vi vaccines have shown efficacy in children living in endemic regions, but a clear correlate of protection is still being established. Vi IgA quantity and avidity, as well as higher fold increases in Vi IgG responses, are correlated with protection from S. Typhi infection, while targeted antibody-mediated functional responses play a key role in the protective signature. These humoral markers can help evaluate and develop effective Vi-conjugate vaccines for typhoid endemic regions.

Summary: This study assessed the emerging antimicrobial resistance (AMR) in pediatric clinical isolates of Salmonella enterica subspecies enterica serovar Typhi and Salmonella enterica subspecies enterica serovar Paratyphi A. The high throughput sequences of twenty pediatric isolates were screened for common antimicrobial resistance genes (ARGs) and virulent factors (VFs). The study identified several prevalent ARGs and observed a correlation between azithromycin resistance and resistance to other antibiotics. The findings will contribute to future studies in designing antibacterial compounds against S. Typhi and S. Paratyphi A.

Summary: This study reveals the production mechanism of Vi antigen. Two proteins, TviE and TviD, encoded in the viAB locus, are identified as Vi antigen polymerase and O-acetyltransferase, respectively. Their functions and mechanisms are determined through structural modeling and mutagenesis experiments. These findings contribute to understanding the production process of Vi antigen and provide foundational information for glycoengineering of Vi antigen production in heterologous bacteria.

Summary: Our study aimed to investigate the effects of deleting tolC on the adhesion and invasion of HT-29 epithelial cells and THP-1 macrophages by S. Typhi in vitro. Our results showed that the tolC mutant strain had significantly reduced adhesion and invasion rates compared to the wild-type strain in both cell types. Additionally, we observed downregulation of SPI-1 gene expression in the tolC mutant.

Summary: In this study, a new serosurveillance tool was applied to estimate the burden of typhoidal Salmonella in a population in Juba, South Sudan. Using dried blood spot testing, the study found an enteric fever seroincidence rate of 30/100 person-years and a cumulative incidence of 74% over a 4-year period.

Summary: Serum salmonellacidal antibody can be used to detect the functional capacity of antibody in patients with enteric fever and after typhoid vaccination. The study found that patients with typhoid and paratyphoid A had significantly higher levels of salmonellacidal antibody titers compared to the control group, and the titers increased significantly with the duration of fever. However, the titers significantly decreased after a certain period of illness. Additionally, although low salmonellacidal titers were detected after Vi CPS vaccine among the human volunteers, the mean titer increased after vaccination.

Summary: Salmonellosis is a major cause of morbidity and mortality, particularly in Latin America where the understanding of mortality rates and antibiotic resistance is limited. This retrospective observational study analyzed patients infected with Salmonella spp. in a university hospital in Colombia between 2012 and 2021. The study found that Salmonellosis accounted for 0.01% of all medical consultations and the most common clinical presentation was gastroenteric syndrome. Mortality occurred in 4.02% of the patients, with higher rates in patients with hematologic malignancy. Antibiotic resistance rates have been steadily increasing over the past 10 years.

Summary: Improvements in household water, sanitation, and hygiene (WASH) as well as typhoid vaccination can reduce typhoid risk in endemic settings. However, the combined impact of these interventions has not been empirically evaluated. This study found that both WASH and ViPS vaccination were associated with reduced typhoid risk, but WASH did not provide additional protection in the ViPS population. These findings highlight the importance of assessing the combination of WASH and typhoid vaccines, and have implications for the evaluation of new-generation typhoid conjugate vaccines.
