Ribavirin reduces mortality in enterovirus 71-infected mice by decreasing viral replication

Enterovirus 71 (EV71) causes fatal encephalitis in young children. However, there is no effective antiviral drug available for infected patients. Ribavirin is currently used for the treatment of several RNA virus infections clinically, so its anti-EV71 efficacy was evaluated. In vitro results showed that ribavirin effectively reduced the viral yields (with an IC50 of 65 mu g/mL) and virus-induced cytopathic effect in human and mouse cell lines. In vivo results showed that ribavirin reduced the mortality, morbidity, and subsequent paralysis sequelae in infected mice by decreasing viral loads in tissues. Thus, ribavirin could be a potential anti-EV71 drug.

Summary: The study showed that Nox2 deficiency attenuates neural damage induced by EAE, reduces disease severity, pathogenic immune cells infiltration, demyelination, and oxidative stress in the CNS. It also revealed that microglial Nox2 plays a regulatory role in multiple pathways important for MS/EAE pathogenesis, suggesting that Nox2 inhibition may represent an important therapeutic target for MS.

Activation of the MAC1-ERK1/2-NOX2 Pathway Is Required for LPS-Induced Sustaining Reactive Microgliosis, Chronic Neuroinflammation and Neurodegeneration

Summary: Recent studies have found that improper resolution of acute neuroinflammation can lead to long-lasting low-grade chronic neuroinflammation and progressive neurodegeneration. This study aimed to identify the potential pathways mediating chronic neuroinflammation and resultant neurodegeneration following LPS stimulation. The results showed that TLR-4 activation, which is essential for acute neuroinflammation, is not sufficient for maintaining chronic neuroinflammation. Instead, the MAC1-NOX2-ERK1/2 signaling pathway plays a crucial role in the initiation and maintenance of low-grade chronic neuroinflammation. Continuous activation of ERK1/2 and NOX2 in microglia forms a vicious cycle, sustaining the low-grade neuroinflammation and driving neurodegeneration.

Complement C3 Enhances LPS-Elicited Neuroinflammation and Neurodegeneration Via the Mac1/NOX2 Pathway

Summary: Recent studies have found increased expression of complements in neurodegenerative diseases like Alzheimer's and Parkinson's diseases. However, the mechanism and role of complements in neurodegeneration are unclear. This study showed that acute neuroinflammation increased the expression of complement C3, and sustained expression during chronic neuroinflammation required the release of damage-associated molecule patterns (DAMPs). The activation of microglial NADPH oxidase (NOX2) was found to be involved in the sustained expression of C3. The findings suggest a role of C3 in regulating chronic neuroinflammation and driving progressive neurodegeneration.
