Inflammatory Cytokines Break Down Intrinsic Immunological Tolerance of Human Primary Keratinocytes to Cytosolic DNA

Keratinocytes are involved in protecting the body from infections and environmental challenges, but also in inflammatory conditions like psoriasis. DNA has emerged as a potent stimulator of innate immune responses, but there is largely no information of how keratinocytes respond to cytosolic DNA. In this study, we report that human keratinocytes are tolerant to cytoplasmic DNA. However, if treated with inflammatory cytokines, keratinocytes gained the capacity to respond to DNA through a mechanism antagonized by the antimicrobial peptide LL37, proposed to be involved in activation and regulation of skin inflammation. The DNA sensor IFN-inducible protein 16 (IFI16) colocalized with DNA and the signaling molecule stimulator of IFN genes (STING) in the cytoplasm only in cytokine-stimulated cells, correlating with recruitment of the essential kinase TANK-binding kinase 1. Moreover, IFI16 was essential for DNA-driven innate immune responses in keratinocytes. Finally, IFI16 was upregulated in psoriasis skin lesions and localized to the cytoplasm in a subpopulation of cells. Collectively, this work suggests that inflammatory environments in the skin can lead to breakdown of tolerance for DNA in keratinocytes, which could contribute to the development of inflammatory diseases.

Summary: Cutaneous T-cell lymphoma (CTCL) is a devastating lymphoid malignancy characterized by the accumulation of malignant T cells in the dermis and epidermis. Skin lesions cause serious symptoms and increase the risk of bacterial infection. The study found that malignant T cells secrete cytokines that downregulate the expression of filaggrin and filaggrin-2, leading to compromised skin barrier. Treatment with JAK inhibitors can reverse this effect and increase filaggrin expression in patients with advanced CTCL.

Guselkumab, tildrakizumab, and risankizumab in a real-world setting: drug survival and effectiveness in the treatment of psoriasis and psoriatic arthritis

Summary: This study retrospectively analyzed the drug survival and effectiveness of interleukin-23 inhibitors in the treatment of psoriasis and psoriatic arthritis in a real-world setting. The results showed that these drugs have high and similar drug survival and effectiveness in patients with difficult-to-treat psoriasis and PsA.

Kidney function before and after acute kidney injury: a nationwide population-based cohort study

Endolysin Inhibits Skin Colonization by Patient-Derived Staphylococcus Aureus and Malignant T-Cell Activation in Cutaneous T-Cell

Summary: This study investigates the effect of a recombinant, antibacterial protein, endolysin (XZ.700), on S. aureus skin colonization and malignant T-cell activation in cutaneous T-cell lymphomas. The results show that endolysin strongly inhibits S. aureus proliferation in the skin of lymphoma patients and reduces bacterial cell counts in a dose-dependent manner. Moreover, endolysin also inhibits the colonization of S. aureus on healthy and lesional skin and has a significant inhibitory effect on the activation and proliferation of malignant T cells induced by S. aureus.

Heat shock protein 90 inhibitor RGRN-305 potently attenuates skin inflammation

Transcriptomic Analysis of Hidradenitis Suppurativa: A Unique Molecular Signature with Broad Immune Activation

Summary: Hidradenitis suppurativa is a chronic inflammatory skin disease with limited treatment options. This study used transcriptomic approaches to investigate the underlying inflammatory pathways and cell types in hidradenitis suppurativa, revealing the activation of multiple inflammatory processes and the involvement of lymphoid and myeloid cells. The study also found increased expression of TH1/2/17 signatures in hidradenitis suppurativa. These findings provide molecular insights into the pathomechanisms of hidradenitis suppurativa and suggest potential therapeutic targets.

An IKBKE variant conferring functional cGAS/STING pathway deficiency and susceptibility to recurrent HSV-2 meningitis

Summary: This study reveals the importance of the IKBKE gene and the cGAS/STING pathway in protecting against recurrent HSV-2 meningitis. A monoallelic variant in the IKBKE gene was identified in a patient with multiple episodes of HSV-2 meningitis, leading to impaired induction of IFN genes and decreased antiviral activity. The patient allele encoded a truncated IKK epsilon protein with loss of kinase activity and dominant-negative activity. Reintroducing wild-type IKBKE into patient cells restored IFN induction and antiviral activity. These findings highlight the nonredundant role of the cGAS/STING pathway in human antiviral immunity.
