Topical Application of Soluble CD83 Induces IDO-Mediated Immune Modulation, Increases Foxp3+ T Cells, and Prolongs Allogeneic Corneal Graft Survival

Modulation of immune responses is one of the main research aims in transplant immunology. In this study, we investigate the local immunomodulatory properties of soluble CD83 (sCD83) at the graft-host interface using the high-risk corneal transplantation model. In this model, which mimics the inflammatory status and the preexisting vascularization of high-risk patients undergoing corneal transplantation, allogeneic donor corneas are transplanted onto sCD83-treated recipient animals. This model allows the direct and precise application of the immune modulator at the transplantation side. Interestingly, sCD83 was able to prolong graft survival after systemic application as well as after topical application, which is therapeutically more relevant. The therapeutic effect was accompanied by an increase in the frequency of regulatory T cells and was mediated by the immune-regulatory enzyme IDO and TGF-beta. In vitro, sCD83 induced long-term IDO expression in both conventional and plasmacytoid dendritic cells via autocrine or paracrine production of TGF-beta, a cytokine previously shown to be an essential mediator of IDO-dependent, long-term tolerance. These findings open new treatment avenues for local immune modulation after organ and tissue transplantation.

Summary: This study compares the long-term outcome of Descemet membrane endothelial keratoplasty (DMEK) in patients with Fuchs endothelial corneal dystrophy (FECD) and pseudophakic bullous keratopathy (PBK). The results suggest that visual acuity after DMEK is comparable in PBK and FECD patients during long-term follow-up, but corneal edema recurs faster in PBK patients.

Summary: The aim of this study was to investigate the influence of dietary supplementation using Age-Related Eye Disease Study 2 (AREDS2) on complement activation in patients with neovascular age-related macular degeneration (nAMD) under ongoing treatment. The results showed that short-term dietary AREDS2 supplementation leads to a significant increase in systemic zinc levels without any influence on complement activation levels.

Summary: In this study, researchers investigated the effect of human amniotic fluid extracellular vesicles (HAF-EVs) on inflammasome activation in monocytes. They found that HAF-EVs expressed immunoregulatory molecules and small amounts of endotoxin. Surprisingly, metagenomic analysis revealed the presence of specific bacterial strain variants associated with HAF-EVs as potential sources of endotoxin. These findings suggest that HAF-EVs may play a role in immune modulation and could serve as a source of specific microbiota during early pregnancy.

Extracellular Vesicles in Aging: An Emerging Hallmark?

Summary: Extracellular vesicles (EVs) are membrane-enclosed particles secreted by cells and circulating in body fluids. They not only serve as a tool for waste disposal, but also transmit cell signals. Aging is characterized by genomic instability, telomere erosion, epigenetic alteration, and defective proteostasis. Studies have shown that EVs released in cell models are associated with these aging hallmarks and can spread senescence signals. Furthermore, EVs have great potential as anti-aging tools. Although this is a recent field, altered EV secretion in aging suggests it as an emerging hallmark of aging.

Summary: Ultraviolet light from the sun causes DNA damage, especially to the skin and eyes of humans. Two repair enzymes from the kangaroo rat and an anti-bacterial virus show potential in repairing UV-induced lesions. While research and commercial products exist for skin repair, there is little research and no available products for eye protection. Our study aims to offer new options for protecting the eyes, which are particularly sensitive to UV rays and require rapid repair.

ABCB5+Limbal Epithelial Stem Cells Inhibit Developmental but Promote Inflammatory (Lymph) Angiogenesis While Preventing Corneal Inflammation

Summary: The limbal epithelial stem cell (LESC) marker ABCB5 was used to investigate its role in corneal neovascularization. It was found that ABCB5 caused an increase in limbal lymphatic and blood vessel complexity in developing mice, but not in adult mice. Furthermore, in an inflammatory challenge, ABCB5 had a pro-lymphangiogenic effect. In addition, ABCB5-positive cells stimulated blood and lymphatic endothelial cell proliferation and migration, and had both pro-(lymph) angiogenic and anti-inflammatory properties.

Amniotic fluid stem cell-derived extracellular vesicles are independent metabolic units capable of modulating inflammasome activation in THP-1 cells

Summary: This study investigated the ability of human amniotic stem cell-derived extracellular vesicles (HASC-EVs) to interfere with inflammasome activation in THP-1 cells. The results showed that HASC-EVs were not internalized or directly interacted with THP-1 cells. HASC-P10 and P100 subsets of HASC-EVs were found to intrinsically produce ATP, which was converted to adenosine by CD73 and CD39 enzymes. Additionally, treatment of THP-1 cells with HASC-EVs prevented the activation of the NLRP3/caspase-1/inflammasome platform, involving the activation of A2a adenosine receptor. These findings suggest that HASC-EVs function as independent metabolic units capable of modulating cellular functions and exerting anti-inflammatory effects in monocytic cells.
