Summary: This study confirmed the immunosuppressive effect of human cord blood MDSCs on corneal allograft models. Local and systemic MDSC administration inhibited the maturation of dendritic cells and the differentiation of Th1 cells, while reducing the infiltration of inflammatory cells and angiogenesis. The results suggest that CB-MDSCs have therapeutic potential for preventing corneal allograft rejection.

Summary: Previous studies have shown that host immunity in paracoccidioidomycosis (PCM) is regulated by tolerogenic plasmacytoid dendritic cells, IDO-1, and Tregs. This study investigated the involvement of MDSCs in PCM and found that they played a role in immunosuppression. MDSCs were found to infiltrate the lungs during Paracoccidioides brasiliensis infection and their reduction resulted in a robust T-cell response and reduced disease severity. The study suggests that MDSCs could be a potential target for adjuvant therapy in PCM.

Summary: Cancer immunotherapy targeting myeloid-derived suppressor cells (MDSCs) is a promising anti-cancer strategy. This study explored the regulatory mechanisms of cholesterol metabolic reprogramming in MDSCs and found that cholesterol deficiency increased the immunosuppressive activity of MDSCs. Mechanistically, cholesterol elimination induced nuclear accumulation of LXR beta and enhanced the binding of LXR beta-RXR alpha on Arg1 promoter. Furthermore, itraconazole improved MDSC immunosuppressive activity and promoted tumor growth by inhibiting the RIPK3-AKT-mTORC1 pathway and impeding cholesterol synthesis.

Summary: This study aimed to investigate the densities and locations of CD11b(+) myeloid-derived suppressor cells (MDSCs) in oral squamous cell carcinoma (OSCC) and determine their prognostic significance. High densities of CD11b(+) MDSCs in the tumor center (CT) and invasive margin (IM) were significantly associated with inferior overall and disease-free survival. CD11b CT and CD11b IM were identified as independent prognostic predictors for patient survival, with higher accuracy and specificity compared to other prognostic parameters.

Summary: This study has demonstrated that exosomes released from rapamycin-conditioned myeloid-derived suppressor cells (Rapa-Exo) significantly prolong the survival of corneal allografts. Mechanistically, exosomal miR-181d-5p directly targets KLF6, resulting in reduced expression of proinflammatory cytokines and ultimately contributing to the resolution of inflammation and improvement of corneal allograft rejection.
