Summary: To address the crisis of Klebsiella pneumoniae infections caused by drug-resistant strains, new approaches to antimicrobial therapy, such as using phages or phage derivatives, need to be developed. This study describes the first K. pneumoniae phage from the Zobellviridae family, which was isolated from river water and demonstrates lytic activity against K. pneumoniae strains with the KL20 capsule type. The phage's receptor-binding protein was identified as a polysaccharide depolymerase, showing promise for using depolymerases in antimicrobial therapy.

Summary: The development of new antimicrobial drugs is necessary to combat the global increase of multi-drug resistant and novel hypervirulent strains of Klebsiella pneumoniae (KPN) associated with higher morbidity and mortality. This study used computer integrated-drug discovery methods and binding-free energy calculations to identify three novel inhibitors (21272541, 67724550, and 67724551) of the FabI protein, which plays a crucial role in fatty acid biosynthesis. All three inhibitors showed strong affinity, with 21272541 being the most effective inhibitor. These inhibitors hold promise for the development of new antimicrobial drugs against antimicrobial resistant KPN infections, pending further in-vitro and in-vivo clinical studies to confirm their efficacy.

Summary: Klebsiella pneumoniae (KP) has caused disease outbreaks in animals, leading to economic losses and biosafety concerns. Vaccines are effective tools against KP infection, but there is currently no KP vaccine available in veterinary medicine. The highly conserved outer membrane phosphoporin (PhoE) of KP has been found to be immunogenic in mice, inducing high levels of specific antibodies and robust cell-mediated immunity. Immunization with PhoE also increased the secretion of IFN-gamma and IL-4, suggesting the induction of mixed Th1 and Th2 responses. Mice immunized with PhoE showed significantly higher complement-mediated lysis of KP cells, and were protected against KP challenge with improved survival and reduced bacterial load.

Summary: In the face of increasing antibiotic-resistant bacteria, it is crucial to find alternatives to conventional antibiotics to combat the serious threat they pose to humanity and the financial burden they impose on communities. This study developed a co-encapsulated formulation of Vancomycin and Gingerol in niosomes and evaluated its effectiveness as an antibacterial agent against carbapenem-resistant Klebsiella pneumoniae strains. The optimized formulation showed sustained drug release, great stability, and enhanced antibacterial and antibiofilm activity against CRKP strains, making it a promising candidate for targeted drug delivery.

Summary: This study found that CD63 remains a stable marker at Day 1 under temperature-controlled conditions, while the CD203c ratio significantly decreases at Day 1. Calculating the CD63/IgE ratio proves to be more stable than CD63 alone. The most stable readouts are the semi-quantitative results and the trajectory of the dose response curve. Ultimately, the stability of basophil activation assay can be extended to 1 day for samples stored at 18-25 degrees C and samples shipped under ambient conditions within the 2-37 degrees C range.

Summary: Our study revealed a distinct expression pattern of caspases in immune cells of COVID-19 patients compared to controls, with upregulation of caspase-1 in CD4+ T cells of hospitalized COVID-19 patients. Post-COVID-19 patients with lingering symptoms also showed increased caspase-1 activity in CD4+ T cells, which was attenuated with a pan-caspase inhibitor in ex vivo experiments. Elevated caspase-3/7 levels in red blood cells from COVID-19 patients were reduced following caspase inhibition.
