Lethal Effect of CD3-Specific Antibody in Mice Deficient in TGF-β1 by Uncontrolled Flu-Like Syndrome

CD3-specific Ab therapy results in a transient, self-limiting, cytokine-associated, flu-like syndrome in experimental animals and in patients, but the underlying mechanism for this spontaneous resolution remains elusive. By using an in vivo model of CD3-specific Ab-induced flu-like syndrome, we show in this paper that a single injection of sublethal dose of the Ab killed all TGF-beta 1(-/-) Mice. The death of TGF-beta 1(-/-) mice was associated with occurrence of this uncontrolled flu-like syndrome, as demonstrated by a sustained storm of systemic inflammatory TNF and IFN-gamma cytokines. We present evidence that deficiency of professional phagocytes to produce TGF-beta 1 after apoptotic T cell clearance may be responsible, together with hypersensitivity of T cells to both activation and apoptosis, for the uncontrolled inflammation. These findings indicate a key role for TGF-beta 1 and phagocytes in protecting the recipients from lethal inflammation and resolving the flu-like syndrome after CD3-specific Ab treatment. Summary: Plasmacytoid dendritic cells (pDCs) in non-small cell lung cancer patients have been shown to have an ambivalent role in cancer immunity. Higher levels of circulating pDCs are associated with improved overall survival (OS) in NSCLC, especially in early-stage patients, and a high pDC signature in the tumor microenvironment predicts better efficacy of immune checkpoint inhibitors in these patients.

Pyrolyzed deketene curcumin controls regulatory T cell generation and gastric cancer metabolism cooperate with 2-deoxy-d-glucose

Summary: Pyrolyzed deketene curcumin GO-Y022 prevents carcinogenesis in a gastric cancer mouse model, but its effect on tumor-induced immune suppression is still unclear. In this study, it was found that GO-Y022 inhibited Treg generation in the presence of transforming growth factor beta 1 (TGF-beta). However, GO-Y022 had limited impact on Foxp3(+) Tregs in the gastric tumor microenvironment. Gastric tumor cells produced a large amount of L-lactate in the presence of GO-Y022, which undermined the inhibitory effect of GO-Y022 on Treg generation in response to TGF-beta. Combination treatment with GO-Y022 and 2-deoxy-d-glucose (2DG) reduced L-lactate production and Treg generation in gastric tumor cells, leading to inhibition of gastric tumor cell survival and promotion of anti-tumor immunity.

Dendritic Cell Subsets in Melanoma: Pathophysiology, Clinical Prognosis and Therapeutic Exploitation

Summary: Immunotherapies using immune checkpoint blockers have shown clinical success in melanoma, but some patients still relapse or do not respond. Dendritic cells (DCs) play a key role in immune responses and their understanding is important for developing effective immunotherapies. This review provides an overview of DC diversity, their role in melanoma pathophysiology and clinical outcomes, tumor-induced DC subversion mechanisms, and DC-based therapeutic developments.

Curcumin analog GO-Y030 inhibits tumor metastasis and glycolysis

Summary: In this study, the antitumor effects of GO-Y030, a curcumin analog, were investigated in a mouse model. The results showed that GO-Y030 treatment inhibited the expression of TGF-β and glycolysis in melanoma cells, leading to the inhibition of tumor invasion and metastasis. Furthermore, GO-Y030 administration demonstrated efficacy in inhibiting lung tumor metastasis without affecting vascular endothelial cells.

Pre-transplant immune profile defined by principal component analysis predicts acute rejection after kidney transplantation

Summary: Unsupervised hierarchical clustering analysis was performed on pre-transplant immunological phenotype in 1113 renal transplant recipients, identifying three immune profiles correlated with clinical phenotypes. The memory immune cluster was associated with older age and diabetes, while the activated immune cluster was associated with younger age and previous exposure to CMV. Both were associated with an increased risk of acute rejection.

The study found that melanoma tumour cells display aberrant glycan motifs which can be sensed by dendritic cells through specific receptors. This interaction may lead to abnormal immune responses. The study suggests that targeting the glycan/receptor axis could be a promising strategy for improving cancer immunotherapy.

Plasma citrulline concentration and plasma LPS detection among critically ill patients a prospective observational study

Summary: The aim of this study was to investigate the correlation between plasma citrulline levels and plasma lipopolysaccharide (LPS) concentration, as well as the impact of sepsis on gut function in critically ill patients. The results showed no correlation between plasma citrulline concentration and plasma LPS concentration or activity. However, septic patients had significantly lower plasma citrulline levels. Additionally, abdominal sepsis was associated with higher plasma LPS activity compared to extra-abdominal sepsis.
