Summary: This study designed an injectable peptide amphiphile (PA) nanofiber that can provide sustained and local losartan release, promoting cartilage regeneration. The results showed that the positively-charged nanofibers not only promoted cell proliferation but also significantly increased the production of glycosaminoglycans in cartilage. Moreover, gene expression analysis suggested that losartan-encapsulated nanofibers had significant anti-inflammatory, anti-degenerative, and cartilage regenerative effects by significantly blocking TGF-beta 1 in this in vitro system.

Summary: In this study, abnormal expression of tissue factor (TF) was found to be associated with poor prognosis and low immune effector cell infiltration in triple-negative breast cancer (TNBC). Inhibition of TF in tumor cells was shown to inhibit tumor growth and increase infiltration of effector T cells. Furthermore, a fusion protein targeting both TF and TGFβ showed enhanced anti-tumor effects and improved tumor immune environment. The study also revealed a novel mechanism of TF in TNBC, which involves blocking the synthesis and secretion of chemokines CXCL9/10/11 to inhibit T cell recruitment and function.

Summary: Vg9Vd2+ T cell-targeted immunotherapy is a promising approach for cancer treatment. This study identified BTN2A1 and BTN3A1 as signal 1 molecules for Vg9Vd2 TCR activation and found that a costimulatory signal is required for optimal activation. Furthermore, a bispecific gd T cell engager BTN2A1/3A1-Fc-CD19scFv was shown to enhance killing of CD19+ lymphoma cells, indicating the importance of costimulatory ligands on tumor cells. These findings highlight the similarities between signal 1 and signal 2 requirements in ab and gd T cell activation and demonstrate the potential of heterodimeric BTNs for targeted activation of gd T cells.

Summary: The ability to produce IL-2 identifies constituents of the expanded CD8 T cell effector pool with stem-like features, which exhibit superior protective powers. The production of IL-2 by cells attenuates the ability to receive IL-2 signals, limiting terminal effector formation and conferring superior protection. Non-producing effector cells, on the other hand, gain effector traits at the expense of memory formation. Despite distinct properties during the effector phase, IL-2-producing and non-producing CD8 T cells converge transcriptionally as memory matures, forming populations with equal recall abilities.

Summary: In a murine model of dry eye disease (DED), a linear transition from effector to memory Th17 cells was detected during the abatement phase of acute inflammation, with persistently high levels of IL-23 and diminished levels of IL-2. Additionally, in vitro culture of effector Th17 cells with IL-23 but not IL-2 led to significant generation of memory Th17 cells. The data demonstrate a new dichotomy between IL-23 and IL-2 in driving effector Th17 cells into the memory pool in autoimmune-mediated ocular surface inflammation.

Summary: Pterostilbene (PTE) has shown anti-tumor effects on glioma cells by inhibiting proliferation, inducing apoptosis, and inhibiting migration through the generation of ROS and reduction of MMP. Activation of ERK 1/2 and JNK signaling pathways contribute to glioma cell apoptosis induced by PTE, which can be attenuated by specific inhibitors of these pathways.
