Indoleamine 2,3-dioxygenase 1 (IDO1) catabolizes tryptophan to kynurenine at the first step of tryptophan metabolism. Recently, in addition to IDO1, a new isoform called IDO2 was identified. In this study, we examined the tissue expression pattern of IDO2 mRNA and the cellular localization of expressed IDO2 protein in mice. IDO1 mRNA expression was observed in the colon and epididymis, whereas IDO2 mRNA expression was found in the cerebral cortex, liver, kidney, and epididymis. Immunohistochemical analysis revealed that IDO2 protein was exclusively expressed on the hepatocytes, interlobular bile ducts, neuronal cells of the cerebrum cortex, Purkinje cells of the cerebellum cortex, lamina epithelialis, proximal convoluted tubule, and the collecting tubule of the kidney. In the epididymis, IDO1 protein expression was restricted to the caput, whereas IDO2 protein expression was observed on the caput, corpus, and cauda. Both IDO proteins were expressed on the caput, but both showed a different protein expression pattern in the segments. Immunohistochemical analysis in IDO1-/- mouse epididymis showed that IDO2 protein was extensively upregulated due to the loss of IDO1 expression. (J Histochem Cytochem 60:854-860, 2012)

Yorkie drives supercompetition by non-autonomous induction of autophagy via bantam microRNA in Drosophila

Rina Nagata, Nanami Akai, Shu Kondo, Kuniaki Saito, Shizue Ohsawa, Tatsushi Igaki

Summary: Mutations in the tumor-suppressor Hippo pathway activate the transcriptional coactivator Yorkie, leading to autonomous cell proliferation and non-autonomous cell death. This study reveals that Yki-induced supercompetition is crucial for tumorigenesis and is driven by non-autonomous induction of autophagy.

Summary: A gain-of-function variant in the GRIA3 gene was found to cause a distinct neurodevelopmental disorder phenotype compared to loss-of-function variants. Drugs that suppress glutamatergic neurotransmission may improve the symptoms of this phenotype. This study is significant for the clinical management of GRIA3-related NDDs.

D-allose enhances the efficacy of hydroxychloroquine against Lewis lung carcinoma cell growth by inducing autophagy

Summary: D-Allose, a rare sugar, can inhibit tumor cell growth by blocking glucose uptake. However, some cells develop resistance to D-Allose. In this study, the researchers established a D-Allose-resistant tumor cell line and a mouse xenograft model. They found that combined treatment with D-Allose and an autophagy inhibitor effectively suppressed tumor growth and selectively induced autophagy at the tumor site.

Summary: The partial loss of liver due to liver transplantation or acute liver failure induces rapid liver regeneration. Recently, we reported that the selective inhibition of indoleamine 2,3-dioxygenase (Ido) 1 promotes early liver regeneration. However, the role of Ido2 in liver regeneration remains unclear.

Summary: In Drosophila, specific splice variants of the insulator protein Mod(mdg4)-N repress HeT-A expression by blocking subtelomeric enhancers and associating with the subtelomeric sequences. Furthermore, Mod(mdg4)-N recruits RNA polymerase II to the subtelomeres, reinforcing its enhancer-blocking function.

Summary: This study proposes an automatic classification method for idiopathic interstitial pneumonia (IIP) using deep learning, and compensates for the lack of data on rare diseases by generating pathological images using a generative adversarial network (GAN). The results show that this method is effective for the diagnosis of idiopathic pulmonary fibrosis (IPF), with high accuracy.

Maternal Inflammation with Elevated Kynurenine Metabolites Is Related to the Risk of Abnormal Brain Development and Behavioral Changes in Autism Spectrum Disorder

Summary: Several studies have shown that genetic and environmental factors play significant roles in the development and progression of neurodevelopmental disorders. Maternal immune activation during pregnancy is considered a major environmental factor in this process. The activation of the kynurenine pathway following neuro-inflammation generates neuroactive metabolites that can affect brain functions and behaviors. Investigating the role of kynurenine pathway metabolites during neurodevelopment can provide valuable insights into the pathophysiology of neurodevelopmental disorders. Evaluating clinical data and animal models, this review examines the impact of perinatal kynurenine pathway elevation on long-term biochemical, neuropathological, and behavioral deficits, with the aim of informing new therapeutic strategies and interventions for these disorders.
