Variation of the gene encoding the nuclear bile salt receptor FXR and gallstone susceptibility in mice and humans

Background/Aims: From quantitative trait locus mapping in inbred mice, we identified the Nr1h4 gene encoding the nuclear bile salt receptor FXR as a candidate gene for the cholesterol gallstone susceptibility locus Lith7. Here, we investigated further an association of the gene encoding FXR and gallstone susceptibility in mice and humans. Methods: The Nr1h4 gene was sequenced in inbred mouse strains with susceptible and resistant Lith7 alleles. Quantitative RT-PCR was employed to determine mRNA expression levels. Gallstone carriers and control subjects of three different populations comprising 1004 individuals were genotyped for polymorphisms of the orthologous human gene detected by sequencing. Results: Expression and sequence analyses in inbred mice were consistent with Nr1h4 underlying Lith7. In the human populations, we identified three frequent haplotypes that accounted for > 95% of all haplotypes observed. In a Mexican population, the most common haplotype NR1H4_1 was associated with gallstone prevalence. In contrast, NR1H4_1 displayed no association with gallstone prevalence in a German population, whereas in a Chilean population we observed a trend towards a protective effect of NR1H4_1. Conclusions: Our study in an inbred mouse model and in three ethnically distinct populations indicates complex interactions of NR1H4 alleles and other risk factors for the development of cholelithiasis. Summary: Despite the ongoing debate, recent studies have shown that under strict contamination-free conditions, various methods combined with advanced bioinformatics tools can detect and quantify bacteria and bacterial DNA in human tissues. Changes in bacterial load and composition in the blood have been associated with several disease states, potentially contributing to systemic inflammation in cardiometabolic disease. This review explores the clinical relevance of a blood bacterial signature to cardiometabolic diseases and proposes strategies for future research and application, considering the challenges posed by low bacterial biomass.

Antifibrotic Soluble Thy-1 Correlates with Renal Dysfunction in Chronic Kidney Disease

Summary: Kidney fibrosis, a major factor in chronic kidney disease (CKD), leads to irreversible organ function loss. This study investigated the association of the soluble form of Thymocyte differentiation antigen-1 (sThy-1) with clinical parameters in CKD patients receiving hemodialysis. The study also examined Thy-1 tissue expression in a mouse model of diabetic CKD. The results showed that sThy-1 levels increased with worsening renal function, regardless of the presence of diabetes. Serum creatinine was found to be a major predictor of sThy-1 levels. Additionally, sThy-1 was found to be an independent predictor of markers of renal function. The study suggests that Thy-1 may have a role in controlling kidney fibrosis and could potentially be used as a renal antifibrotic factor.

The serpin vaspin has compensatory roles in inflammation and insulin resistance. A new transgenic mouse line with human vaspin expression in adipose tissue has been developed, which showed improved metabolic parameters and increased energy expenditure under high-fat diet conditions.

Adipokines as Clinically Relevant Therapeutic Targets in Obesity

Summary: Adipokines play a crucial role in the development of obesity and its associated metabolic and cardiovascular complications. While some adipokines have been identified and studied, many regulatory mechanisms remain unclear, hindering their clinical application. However, due to their complex metabolic properties, adipokines hold great potential as biomarkers and treatment strategies for obesity and related disorders. This review summarizes current clinical studies on adipokines and their potential diagnostic and therapeutic use in obesity and metabolic disorders, highlighting the promising clinical application of fibroblast growth factors (FGF)-19, FGF-21, and leptin.
