To help determine whether the typical arthropod arrangement was a synapomorphy for the whole Tettigoniidae, we sequenced the mitochondrial genome (mitogenome) of the quiet-calling katydids, Xizicus fascipes (Orthoptera: Tettigoniidae: Meconematinae). The 16,166-bp nucleotide sequences of X. fascipes mitogenome contains the typical gene content, gene order, base composition, and codon usage found in arthropod mitogenomes. As a whole, the X. fascipes mitogenome contains a lower A+T content (70.2%) found in the complete orthopteran mitogenomes determined to date. All protein-coding genes started with a typical ATN codon. Ten of the 13 protein-coding genes have a complete termination codon, but the remaining three genes (COIII, ND5 and ND4) terminate with incomplete T. All tRNAs have the typical clover-leaf structure of mitogenome tRNA, except for tRNA (Ser(AGN)), in which lengthened anticodon stem (9 bp) with a bulged nuleotide in the middle, an unusual T-stem (6 bp in constrast to the normal 5 bp), a mini DHU arm (2 bp) and no connector nucleotides. In the A+T-rich region, two (TA)n conserved blocks that were previously described in Ensifera and two 150-bp tandem repeats plus a partial copy of the composed at 61 bp of the beginning were present. Phylogenetic analysis found: i) the monophyly of Conocephalinae was interrupted by Elimaea cheni from Phaneropterinae; and ii) Meconematinae was the most basal group among these five subfamilies.

The binary combined toxicity of lithium, lead, and manganese on the proliferation of murine neural stem cells using two different models

Huan Luo, Jiayi Li, Bo Song, Bing Zhang, Yixi Li, Zhijun Zhou, Xiuli Chang

Summary: This study explored the toxicity of lithium, lead, and manganese on neural stem cell proliferation, as well as their combined effects. The results showed that the combination of lead and manganese had a synergistic toxicity effect in both models. In the dose-additivity method, mixtures with a high ratio of lithium exhibited synergistic effects, while mixtures with a lower or equal lithium ratio showed antagonistic effects. Additionally, the interactions of lithium and manganese mixtures varied across different endpoints.

Single-cell transcriptomic analysis reveals the adverse effects of cadmium on the trajectory of neuronal maturation

Bo Song, Yuwei Zhang, Guiya Xiong, Huan Luo, Bing Zhang, Yixi Li, Zhibin Wang, Zhijun Zhou, Xiuli Chang

Summary: Cadmium (Cd) is an environmental pollutant that has adverse effects on neuronal maturation. This study used single-cell RNA sequencing to uncover the molecular mechanisms of Cd on neuronal lineage cells. The results showed that Cd alters the trajectory of neuronal maturation, induces differential transcriptome expression, and affects oxidative damage and cell cycle phases. In addition, Cd downregulates the autocrine and paracrine signals of Wnt5a. Activation of Wnt5a effectively rescues neuronal development and promotes maturation.

Pressure transient analysis of multistage fractured horizontal wells based on detailed characterization of stimulated area

Huzhen Wang, Tiancheng Liu, Zhuangzhuang Sun, Chunyao Wang, Yangang Zhang, Yulin Chen, Zhenyu Liu, Zhijun Zhou

Summary: In ultralow permeability tight oil reservoirs, the fracturing scale of multistage fractured horizontal wells (MFHWs) is relatively large, and the artificial fracture system is generally more complex. Numerical methods have unique advantages in characterizing the stimulated area in detail. A numerical model was established to study the influence of the width and permeability of the stimulated area on the pressure response of MFHWs. The analysis shows that the conductivity of the main fractures plays a crucial role in the transient pressure response and development of MFHWs.

Paraquat-induced neurogenesis abnormalities via Drp1-mediated mitochondrial fission

Bing Zhang, Yuwei Zhang, Zhenzi Zuo, Guiya Xiong, Huan Luo, Bo Song, Lina Zhao, Zhijun Zhou, Xiuli Chang

Summary: Neurogenesis is a vital process in the nervous system development, and it is regulated by mitochondrial dynamics. Imbalanced mitochondrial dynamics caused by oxidative stress have been associated with various neurological disorders. In this study, we examined the effects of PQ, a neurotoxic agricultural chemical, on neurogenesis in primary murine neural progenitor cells. Our results showed that PQ inhibited cell proliferation, migration, and neuronal differentiation in a concentration-dependent manner. PQ induced excessive mitochondrial fragmentation and altered the expression of proteins involved in mitochondrial dynamics. Inhibiting mitochondrial fission and using a mitochondria-targeted antioxidant could mitigate the adverse effects of PQ on neurogenesis. These findings suggest that mitochondrial fission could be a potential therapeutic target for PQ-induced neurotoxicity.

A Differential Difference Amplifier Employing Pseudo-Differential CMFB for Neural Signal Recording Applications

Zhijun Zhou, Keping Wang, Longbin Zhu, Rui Yang, Jihong Li, Wenjie Wang, Yuhao Xu, Zhigong Wang

Summary: This brief introduces a differential difference amplifier (DDA) with a pseudo-differential (PD) common-mode feedback (CMFB) for neural signal recording. The DDA includes two input transconductors, one connected to the target neural signal for high input impedance, and another employing a self-stabilized topology for stability and cutoff frequency determination. The use of PD terminal in CMFB increases the common mode rejection ratio (CMRR) by a factor of differential-mode gain without affecting voltage headroom and dynamic range. This PD CMFB is suitable for multi-channel neural signal acquisition, with higher CMRR for more channels. The proposed DDA, designed in a 0.18-mu m CMOS technology, achieves high input impedance, high CMRR, and low power consumption.
