Proteasome inhibitors promote the sequestration of PrPSc into aggresomes within the cytosol of prion-infected CAD neuronal cells

Dysfunction of the endoplasmic reticulum associated protein degradation/proteasome system is believed to contribute to the initiation or aggravation of neurodegenerative disorders associated with protein misfolding, and there is some evidence to suggest that proteasome dysfunctions might be implicated in prion disease. This study investigated the effect of proteasome inhibitors on the biogenesis of both the cellular (PrPC) and abnormal (PrPSc) forms of prion protein in CAD neuronal cells, a newly introduced prion cell system. In uninfected cells, proteasome impairment altered the intracellular distribution of PrPC, leading to a strong accumulation in the Golgi apparatus. Moreover, a detergent-insoluble and weakly protease-resistant PrP species of 26 kDa, termed Prp(26K), accumulated in the cells, whether they were prion-infected or not. However, no evidence was found that, in infected cells, this Prp26K species converts into the highly proteinase K-resistant PrPSc. In the infected cultures, proteasome inhibition caused an increased intracellular aggregation of PrPSc that was deposited into large aggresomes. These findings strengthen the view that, in neuronal cells expressing wild-type PrPC from the natural promoter, proteasomal impairment may affect both the process of PrPC biosynthesis and the subcellular sites of PrPSc accumulation, despite the fact that these two effects could essentially be disconnected.

Scrapie infection and endogenous retroviral expression in sheep lymphoid tissues

Summary: Transmissible spongiform encephalopathies, also known as prion diseases, are fatal neurodegenerative diseases that affect both humans and animals. The study suggests that tissue or cell-specific receptors or cofactors may play a role in controlling differential susceptibility to infection. While endogenous retroviruses (ERV) could potentially be one of these cofactors, the study did not find specific differential expression of OERV in lymphoid tissues following scrapie infection in sheep, unlike previous studies in prion-infected mice and macaques, highlighting the complex interactions involved in prion disease pathogenesis.

Preclinical transmission of prions by blood transfusion is influenced by donor genotype and route of infection

Summary: Variant Creutzfeldt-Jakob disease (vCJD) is a human prion disease resulting from zoonotic transmission of bovine spongiform encephalopathy (BSE), and blood transfusion has been documented as a route of transmission. Leucodepletion is an important measure to reduce the risk of transmission, but it does not completely prevent infection. This study using a BSE-infected sheep model found that red blood cells, platelets, and plasma can all transmit prion infection, and the route and method of infection significantly affect the time course of detection in blood, which has implications for disease control.

Summary: Subclinical infections of prion diseases play an important role in the transmission and re-emergence of these diseases. A study using a sheep model found that many sheep survived for a long time without showing clinical signs after being exposed to infected blood transfusion. However, low levels of infection were still detectable in lymph nodes and other tissues.
