Comparison of spermatozoa parameters, fine structures, and energy-related factors among tetraploid, hyper-tetraploid, and hyper-triploid loaches (Misgurnus anguillicaudatus)

To evaluate the influence of ploidy elevation and aneuploidy on spermatozoa in the loach Misgurnus anguillicaudatus, we investigated some parameters (motility, concentration, and viability), fine structures (gross morphology, head size, and flagellum length), and energy-related biochemical factors (volume of mitochondrial mass per cell and ATP content) in diploid, hyper-diploid, and hexaploid-range spermatozoa produced in natural tetraploid, hyper-tetraploid, and hyper-triploid male loaches, respectively. Diploid spermatozoa exhibited vigorous movement and sufficient duration of motility similar to those in haploid spermatozoa. They had longer flagella, higher numbers and larger volume of mitochondria, and higher ATP content than haploid spermatozoa of wild-type diploids. No differences were observed in parameters and morphological characteristics between diploid and hyper-diploid spermatozoa. In contrast, the hexaploid-range spermatozoa of hyper-triploid males exhibited poor progressive motility in spite of a higher ATP content of spermatozoa. Spermatozoa with no flagella (36.0%) or multiple flagella (18.6%) were also observed in hyper-triploids. Ratios of head to flagellum length in hexaploid-range spermatozoa were significantly different from those of haploid spermatozoa. In addition to the normal 9 + 2 microtubule structure of the flagellum, an abnormal 9 + 1 microtubule structure was also observed in the spermatozoa of hyper-triploids. J. Exp. Cold-shock androgenesis in common carp (Cyprinus carpio)

Summary: Methods of induction of uniparental inheritance have been developed for model organisms as well as for species of commercial interest, with cold-shock androgenesis showing promise in successfully inducing haploid progeny in common carp. Compared to other techniques, cold-shock androgenesis was found to be effective in common carp, providing a new possibility for isogenic line production.

Summary: The study provides a detailed overview of dentition development in the sterlet sturgeon, revealing dynamic changes in tooth composition at different stages of the fish's life. Early loss of oral teeth and subsequent development of palatal and pharyngeal teeth result in a translocation of functional dentition within the oropharyngeal cavity, ultimately leading to the loss of all teeth. These unique dynamics in sturgeons offer valuable insights into the developmental and evolutionary mechanisms of vertebrate odontogenesis.

Efficient CRISPR Mutagenesis in Sturgeon Demonstrates Its Utility in Large, Slow-Maturing Vertebrates

Summary: In the last decade, CRISPR/Cas9 has been proven to be an effective and efficient method for targeted mutagenesis in genetic model systems. Its high efficiency allows the immediate generation of near-null mosaic mutants, making it valuable for organisms with life histories that preclude the establishment of purebred genetic lines. In the study, the successful use of CRISPR/Cas9 mutagenesis in a non-teleost fish, sterlet Acipenser ruthenus, was reported, indicating the potential for applying this technology to other long-lived, slow-maturing aquatic vertebrates.

Summary: The developmental characteristics of sturgeon embryos are similar to those of African clawed frogs. The role of the vegetal pole (VP) in sturgeon embryos is mainly to produce primordial germ cells, while the rest of the cells serve as cellular yolk. The study suggests that meroblastic cleavage in sturgeons might have evolved through the fusion of vegetal blastomeres.

Summary: Interspecific hybridization may lead to a transition from sexual reproduction to asexual reproduction, but the reasons for this change are not well understood. In this study, we investigated the triggers of asexual reproduction and found that it is directly influenced by the hybrid genetic constitution of the cell. Additionally, the execution of asexual development is regulated by signals from the surrounding gonadal environment and somatic factors.

Comparison of RNA localization during oogenesis within Acipenser ruthenus and Xenopus laevis

Summary: The localization of biomolecules within the oocyte drives the spatial differentiation of daughter cells during embryogenesis. While the localization pathways of vegetal maternal transcripts have been well-described, limited information is available about the localization of animal pole transcripts. In this study, TOMO-Seq analysis on two different species was performed, revealing temporal preferences for transcript localization establishment and the presence of both conserved and species-specific transcripts. Additionally, active remodeling of the maternal RNA pool through de novo transcription and degradation was observed.

Optimization of in vitro culture conditions of common carp germ cells for purpose of surrogate production

Summary: This study optimized the cultivation conditions for germ cells in common carp, and the results showed that using hESC media and RTG2 cell line as feeder cells promoted germ cell proliferation and survival. Additionally, opening the carp from the dorsal side minimized the risk of contamination. These findings are important for common carp breeding, genetic resource conservation, and development of specific techniques.

Vitrification of the ovarian tissue in sturgeons

Summary: The aim of this study was to test the efficacy of needle-immersed vitrification (NIV) for preserving oogonia (OOG) in sterlet and Russian sturgeon ovarian tissue in order to enhance conservation efforts for these endangered fish species. The study found that vitrification of the ovarian tissue using specific solutions resulted in viable OOGs, which were able to successfully colonize recipient gonads after transplantation. The simplicity and practicality of the vitrification procedure make it a valuable tool for conservation work in the field.

Intracytoplasmic sperm injection in sturgeon species: A promising reproductive technology of selected genitors

Does dietary exposure to 17?-ethinylestradiol alter biomarkers related with endocrine disruption and oxidative stress in the adult triploid of Danio rerio?

Summary: This study investigated the comprehensive effects of 17 alpha-ethinylestradiol (EE2) in zebrafish, focusing on endocrine disruption, oxidative stress, and detoxification processes. Adult male triploid zebrafish were exposed to two concentrations of EE2 in their feed for six weeks. The high concentration of EE2 caused extensive mortality and significant changes in reproductive disorders, gonad histology, vitellogenin level, and selected gene expression. The environmentally relevant concentration of EE2 showed no significant differences in vitellogenin but did affect antioxidant and detoxifying enzymes. This is the first study to report the effects of subchronic oral exposure to EE2 in adult triploid zebrafish.
