RNA editing in RHOQ promotes invasion potential in colorectal cancer

RNA editing can increase RNA sequence variation without altering the DNA sequence. By comparing whole-genome and transcriptome sequence data of a rectal cancer, we found novel tumor-associated increase of RNA editing in ras homologue family member Q (RHOQ) transcripts. The adenosine-to-inosine (A-to-I) editing results in substitution of asparagine with serine at residue 136. We observed a higher level of the RHOQ RNA editing in tumor compared with normal tissue in colorectal cancer (CRC). The degree of RNA editing was associated with RhoQ protein activity in CRC cancer cell lines. RhoQ N136S amino acid substitution increased RhoQ activity, actin cytoskeletal reorganization, and invasion potential. KRAS mutation further increased the invasion potential of RhoQ N136S in vitro. Among CRC patients, recurrence was more frequently observed in patients with tumors having edited RHOQ transcripts and mutations in the KRAS gene. In summary, we show that RNA editing is another mechanism of sequence alteration that contributes to CRC progression.

Radiation Response Prediction Model Based on Integrated Clinical and Genomic Data Analysis

Summary: The value of genomic profiling for predicting radiation therapy response was evaluated and a Bayesian network model was constructed. Alterations of DNA repair pathway and smoking somatic signature were associated with durable radiation response. The Bayesian network model provided helpful insights for high precision radiotherapy. However, further validation in prospective cohorts is needed for individualized treatment.

Calsequestrin 2 overexpression in breast cancer increases tumorigenesis and metastasis by modulating the tumor microenvironment

Summary: CASQ2, a newly identified breast cancer-related gene, has been found to potentially regulate tumor-microenvironment interactions by promoting a more aggressive phenotype and altering epithelial-mesenchymal transition (EMT) markers in breast cancer cells. Overexpression of CASQ2 also induces cancer-associated fibroblast characteristics and increases hypoxia-inducible factor 1 alpha (HIF1 alpha) expression in stromal fibroblasts, leading to accelerated tumorigenesis, collagen structure remodeling, and distant metastasis in vivo. CASQ2 may confer more metaplastic features to triple-negative breast cancer cells, suggesting its key role in breast cancer tumorigenesis and metastasis through modulating various aspects of tumor-microenvironment interactions.

Summary: The study evaluated misalignment errors and exonic multi-mapping events in mixed human and mouse single-cell data analysis, and developed a new bioinformatics pipeline to minimize errors. Results showed that the clustering effect of data processed using this pipeline was good compared to unmixed data, effectively identified false-positive signals caused by ambient RNA, and successfully applied to the analysis of multispecies multisample single-cell libraries.

Prognostic Impact of Extramural Lymphatic, Vascular, and Perineural Invasion in Stage II Colon Cancer: A Comparison With Intramural Invasion

Summary: This study aimed to determine whether the location of lymphatic invasion, vascular invasion, and perineural invasion could affect the survival of patients with stage II colon cancer. The results showed that extramural invasion was associated with a worse prognosis, while intramural invasion was not. Therefore, pathologic reports about the location of invasion may be helpful for predicting prognosis and determining the need for adjuvant chemotherapy in stage II colon cancers.

Summary: This study developed a machine learning-based model to identify preoperative multiparametric MR imaging markers associated with prognosis in glioblastoma patients. The model found that nonenhancing T2 high-signal-intensity lesions with the relatively low normalized CBV, low volume transfer constant values, and the lowest ADC values could serve as useful prognostic imaging markers for predicting survival outcomes in patients with glioblastoma.

Summary: The importance of molecular pathology tests has increased in the past decade, leading to a demand for efficient training in this field. The Korean Society of Pathologists established a task force to develop a refined educational curriculum in molecular pathology. An online educational session was conducted, and the understanding of the participants significantly increased in various learning objectives, particularly in the field of next-generation sequencing. The feedback received on the refined curriculum was predominantly positive.
