Herpesvirus entry mediator (TNFRSF14) regulates the persistence of T helper memory cell populations

Memory T helper cells (Th cells) play an important role in host defense against pathogens but also contribute to the pathogenesis of inflammatory disorders. We found that a soluble decoy lymphotoxin beta receptor (LT-beta R)-Fc, which can block tumor necrosis factor (TNF)-related ligands LIGHT (TNFSF14) and LT-alpha beta binding to the herpesvirus entry mediator (HVEM) and the LT-beta R, inhibited the accumulation of memory Th2 cells after antigen encounter and correspondingly reduced inflammatory responses in vivo. Showing that this was a function of the receptor for LIGHT, antigen-specific memory CD4 T cells deficient in HVEM were also unable to persist, despite having a normal immediate response to recall antigen. HVEM-/- memory Th2 cells displayed reduced activity of PKB (protein kinase B; Akt), and constitutively active Akt rescued their survival and restored strong inflammation after antigen rechallenge. This was not restricted to Th2 memory cells as HVEM-deficient Th1 memory cells were also impaired in surviving after encounter with recall antigen. Furthermore, the absence of LIGHT on T cells recapitulated the defect seen with the absence of HVEM, suggesting that activated T cells communicate through LIGHT-HVEM interactions. Collectively, our results demonstrate a critical role of HVEM signals in the persistence of large pools of memory CD4 T cells.

Quantification and Surface Localization of the Hemolysin A Type I Secretion System at the Endogenous Level and under Conditions of Overexpression

Summary: Secretion systems play a vital role in Gram-negative bacteria by allowing communication with the outside world. This study focused on the hemolysin A (HlyA) type I secretion system (T1SS) in Escherichia coli and analyzed its localization and clustering. The results showed that the amount of ToIC, a component of the T1SS, was not affected by overexpression of the HlyBD complex. Superresolution microscopy revealed that the HlyA T1SS did not exhibit polarization and the size of T1SS clusters did not correlate with outer membrane islands.

In Vitro Biological Activity of Natural Products from the Endophytic Fungus Paraboeremia selaginellae against Toxoplasma gondii

Summary: Toxoplasma gondii is a pathogen that can infect various warm-blooded animals, including humans, causing toxoplasmosis. Current treatments for toxoplasmosis have severe side effects and are not effective in eradicating chronic infection. Therefore, there is an urgent need to develop novel anti-toxoplasma drugs with low toxicity. In this study, eight natural products were isolated from the crude extract of the endophytic fungus Paraboeremia selaginellae obtained from the leaves of the plant Philodendron monstera. Six of these natural products showed antitoxoplasma activity with low or no cytotoxicity in human cell lines. These findings suggest that biphenyl ethers, bioxanthracenes, and 5S,6S-phomalactone from P. selaginellae are potential candidates for novel anti-toxoplasma drugs.
