The majority of mutations occur within a 1 kb region downstream of the J gene, regardless of the specific J gene used. Summary: In this study, the importance of PHRF1 in IgA switching was investigated in CH12F3-2A cells and CD19-Cre mice. The results showed that the loss of PHRF1 decreased IgA production and the levels of PARP1, NELF-A, and NELF-D. Interestingly, IgA switching was not significantly reduced in PHRF1 deficient splenic B lymphocytes.

Summary: AID catalyzes the conversion of deoxycytidines to deoxyuracils in immunoglobulin genes to induce somatic hypermutation and class-switch recombination. FAM72A is identified as a major determinant for the error-prone processing of deoxyuracils, enabling AID to exert its full effects on antibody maturation by disabling efficient DNA repair pathways. Overexpression of FAM72A in cancer could promote mutagenesis.

Summary: Activation-induced deaminase (AID) deaminates cytosine within single-stranded DNA. This study found that AID deamination on duplex DNA substrates is reduced when RNase A is added during transcription. The researchers also discovered that AID acts with similar efficiency on RNA-DNA fusion substrates and DNA-only substrates. Based on these findings, they proposed a model in which the mRNA tail plays a critical role in AID loading and collision with DNA strands.

Summary: Research has identified compounds that inhibit AID, potentially controlling antibody diversity and B cell lymphoma development. These compounds share similar chemical structures, suggesting a common mechanism of action. It was also found that AID is continuously expressed for several days after B cell activation to induce class switch recombination.
