The transition from vegetative growth to reproductive development is a complex process that requires an integrated response to multiple environmental cues and endogenous signals. In Arabidopsis thaliana, which has a facultative requirement for vernalization and long days, the genes of the autonomous pathway function as floral promoters by repressing the central repressor and vernalization-regulatory gene FLC. Environmental regulation by seasonal changes in daylength is under control of the photoperiod pathway and its key gene CO. The root and leaf crop species Beta vulgaris in the caryophyllid clade of core eudicots, which is only very distantly related to Arabidopsis, is an obligate long-day plant and includes forms with or without vernalization requirement. FLC and CO homologues with related functions in beet have been identified, but the presence of autonomous pathway genes which function in parallel to the vernalization and photoperiod pathways has not yet been reported. Here, this begins to be addressed by the identification and genetic mapping of full-length homologues of the RNA-regulatory gene FLK and the chromatin-regulatory genes FVE, LD, and LDL1. When overexpressed in A. thaliana, BvFLK accelerates bolting in the Col-0 background and fully complements the late-bolting phenotype of an flk mutant through repression of FLC. In contrast, complementation analysis of BvFVE1 and the presence of a putative paralogue in beet suggest evolutionary divergence of FVE homologues. It is further shown that BvFVE1, unlike FVE in Arabidopsis, is under circadian clock control. Together, the data provide first evidence for evolutionary conservation of components of the autonomous pathway in B. vulgaris, while also suggesting divergence or subfunctionalization of one gene. The results are likely to be of broader relevance because B. vulgaris expands the spectrum of evolutionarily diverse species which are subject to differential developmental and/or environmental regulation of floral transition.

Comparative transcriptomic and epigenetic analyses reveal conserved and divergent regulatory pathways in barley response to temperature stresses

Bo Li, Caixian Yang, Bingzhuang An, Hongpan Wang, Mohammed Albaqami, Salah Fatouh Abou-Elwafa, Le Xu, Yanhao Xu

Summary: This study investigated the epigenetic regulation responses to temperature stress in barley. It was found that high temperature stress had greater deleterious effects on barley seedlings' growth compared to mild low temperature stress. High temperature stress caused more DNA demethylation, and higher levels of histone modifications were observed. Transcription factors played important roles in both mild low temperature and high temperature stress.

Summary: A nuclear pore complex, ZmNUP58, associated with drought and salt tolerance phenotype has been identified, and its overexpression in maize significantly enhances chlorophyll content and antioxidant enzyme activities under stress conditions. RNA-Seq analysis revealed that ZmNUP58 can regulate genes related to phytohormones, osmotic adjustment substances, antioxidant enzymes, and other stress response pathways.

Functional analysis of a late embryogenesis abundant protein ZmNHL1 in maize under drought stress

Guorui Wang, Huihui Su, Salah Fatouh Abou-Elwafa, Pengyu Zhang, Liru Cao, Jiaxu Fu, Xiaowen Xie, Lixia Ku, Pengfei Wen, Tongchao Wang, Li Wei

Summary: Maize is an important crop for global food security, and developing drought-tolerant genotypes is crucial. Through transcriptomic analysis, we identified a drought-responsive gene, ZmNHL1, which belongs to the LEA-2 protein family. Overexpression of ZmNHL1 in transgenic plants improved ROS scavenging and cell membrane permeability, enhancing maize tolerance to drought stress.

Application of Beet Sugar Byproducts Improves Maize Growth and Salt Redistribution in Saline Soils

Saif F. Alharbi, Fahad S. Alotaibi, Lixia Ku, Wenying Zhang, Salah Fatouh Abou-Elwafa

Summary: Maize, a key component of global food security, faces threats from abiotic stresses such as salinity. An experiment was conducted to evaluate the effects of using beet sugar FC treated with phosphoric and sulfuric acids (TFC) and molasses on a maize cultivar in saline soil. The application of 5 tons of TFC and 125 liters of molasses significantly increased plant height and fresh and dry weights of the maize plants, while reducing sodium accumulation and increasing calcium and potassium accumulation. Applying TFC and molasses also improved soil properties and reduced economic costs. TFC shows promise as an organic amendment for saline soils to enhance plant growth and biofortification.

Zhimin Li, Fengran Gao, Yajing Liu, Salah Fatouh Abou-Elwafa, Junlong Qi, Haibo Pan, Xiaomeng Hu, Zhenzhen Ren, Haixia Zeng, Zhixue Liu, Dongling Zhang, Zhangying Xi, Tianxue Liu, Yanhui Chen, Huihui Su, Shuping Xiong, Lixia Ku

Summary: In this study, it was found that the zmgi2 mutant in maize flowered earlier than the wild type under long day conditions. By using DAP-Seq and RNA-Seq, it was further revealed that ZmGI2 delays flowering by repressing the expression of ZmVOZs, ZmZCN8, and ZmFPF1, and promoting the expression of ZmARR11, ZmDOF, and ZmUBC11. This study provides insights into the potential role of ZmGI2 in regulating the flowering time-dependent photoperiodic pathway.

Summary: Traditional breeding is an important method for developing drought-tolerant varieties, and the study of traditional breeding methods is also essential for disease resistance in crops. This research compared the effects of single trait selection and independent culling levels on cotton yield, and found that both methods played a positive role in improving cotton yield. Furthermore, the independent culling level selection method better preserved genetic variability.
