CELLFOOD™ induces apoptosis in human mesothelioma and colorectal cancer cells by modulating p53, c-myc and pAkt signaling pathways

Background: CELLFOOD (TM) (CF) is a nutraceutical non-addictive, non-invasive, and completely non-toxic unique proprietary colloidal-ionic formula. Little is known about its effect on cancer cells in solid tumors. The aim of this study was to evaluate the effect that CF has on different cancer cell lines and the mechanism by which the nutraceutical works. Methods: The effect of CF on HFF (normal fibroblasts), Met5A (mesothelium), MSTO-211H, NCI-2452, Ist-Mes1, MPP89, Ist-Mes2 (mesothelioma), M14 (melanoma), H1650, H1975 (lung cancer), SKRB3 (breast cancer), and HCT-116 (colorectal cancer) cell growth was tested by cell proliferation and clonogenic assay. Among all of them, MSTO-211 and HCT-116 were analyzed for cell cycle by flow cytometry and western blot. Results: All human cancer lines were suppressed on cell growth upon 1: 200 CF treatment for 24 and 48 hours. Death was not observed in HFF and Met5A cell lines. Cell cycle analysis showed an increased sub-G1 with reduction of G1 in MSTO-211 and a cell cycle arrest of in G1 in HCT116. Activation of caspase-3 and cleavage of PARP confirmed an apoptotic death for both cell lines. Increased expression levels of p53, p21, and p27, downregulation of c-myc and Bcl-2, and inhibition of Akt activation were also found in CF-treated MSTO-211 and HCT-116 cells. Conclusions: These findings ascertained an interaction between p53, c-myc, p21, p27, Bcl-2, PI3K/Akt pathway, and CF-induced apoptosis in MSTO-211H and HCT-116 cells, suggesting that CF acts as an important regulator of cell growth in human cancer cell lines. CF could be a useful nutraceutical intervention for prevention in colon cancer and mesothelioma.

Serum changes in sTWEAK and its scavenger receptor sCD163 in ultramarathon athletes running the 24-h race

Summary: In this study, significant decrease of sTWEAK levels and increase of sCD163 levels were observed in the serum of ultramarathon athletes after the race. Positive relationships were found between IL-6 and sCD163 levels with cardiac damage markers, while sTWEAK showed an inverse correlation with IL-6 and NT-proBNP.

Summary: The human amniotic membrane (HAM) has biological properties that promote wound healing, making it an ideal biological dressing for treating clinical conditions such as burns and non-healing wounds. However, preservation methods are necessary to maintain the tissue properties of HAM during banking. Cryopreservation and freezing, commonly used methods, can cause oxidative stress-induced cell injury and affect tissue regenerative properties. This study explores the use of quercetin, a natural compound with antioxidant activity, to improve the antioxidant and regenerative properties of frozen or cryopreserved HAM tissues.

A microplate-based DCFH-DA assay for the evaluation of oxidative stress in whole semen

Summary: This study aimed to develop an assay for reactive oxygen species detection in semen samples. The microplate-based 2',7'-dichlorofluorescein diacetate assay was successfully adapted and showed potential for evaluating oxidative stress in whole semen. It could be used for preliminary assessment of semen oxidation levels and for evaluating the effect of pro-glutathione molecules on semen oxidative status.

Synthesis and Biological Evaluation of 6-O-Sucrose Monoester Glycolipids as Possible New Antifungal Agents

Summary: A library of 6-O-sucrose monoester surfactants was synthesized and tested for their antimicrobial activities. The synthetic procedure, a modified Mitsunobu reaction, showed improved results compared to the literature. Most of these glycolipids exhibited satisfactory antifungal activities, with sucrose palmitoleate (URB1537) showing good activity against Candida albicans ATCC 10231, Fusarium spp., and Aspergillus fumigatus IDRAH01. URB1537 was further characterized for its radical scavenging, anti-inflammatory, and biocompatibility properties, demonstrating control of inflammatory response and safety.

Summary: In this study, we used bioinformatics analysis and experimental validation to investigate the mechanism behind the antioxidant effects of Prunus spinosa fruit (PSF) extract during a pro-inflammatory TLR4 insult. The analysis suggested that the extract activates the biological process of cellular senescence and identified potential targets in ARE signaling, MMP, and p53 SR pathways. Experimental validation showed that the extract reduces oxidative stress through the modulation of Nrf2, but no significant effects were observed on p53 and MMP modulation.

The drug delivery potential and cytocompatibility of these nanoparticles have been evaluated through various experimental measurements.
