Modulation of LPS-stimulated neuroinflammation in BV-2 microglia by Gastrodia elata: 4-Hydroxybenzyl alcohol is the bioactive candidate

Ethnopharmacological relevance: Gastrodia elata Blume (Orchidaceae, GE) a traditional plant in Oriental countries is known for its enormous benefits to treat headaches, dizziness, vertigo and convulsive illnesses. In the present study, the ethnopharmacological role of GE in neuroinflammation mediated by activated microglia and the mechanisms underlying were reported. Materials and methods: BV-2 microglia activated by lipopolysaccharide (LPS) was employed and the effects of GE on corresponding neuroinflammatory parameters were assessed. Results: GE extract inhibited LPS-stimulated production of inflammatory cytokines and down regulated the c-Jun NH2-Terminal Kinase (JNK) and nuclear factor-kappa B (NF-kappa B) signaling pathways, which are known to be involved in neuroinflammation. Further, inhibition of NO and iNOS by 4-hydroxybenzyl alcohol (4-HBA), one of the active constituent of GE in LPS-stimulated BV-2 cells suggest that 4-HBA might be the bioactive candidate. Conclusion: GE extract and its active constituent 4-HBA could be further exploited to mitigate microglial activation and may be developed as a new therapeutic remedy in treating various neuroinflammatory diseases. Summary: Piperine (PIP) has shown potential therapeutic effects in various diseases, including inflammation, cardiac and hepatic diseases, neurodegenerative diseases, and cancer. Although PIP has poor pharmacokinetic properties, its interaction with metabolic enzymes and the Monoamine oxide B (MAO-B) active site make it a xenobiotics bioenhancer and a potential MAO-B inhibitor. Advancements in pharmaceutical technology have improved PIP's limitations. However, the structure activity relationship (SAR) study suggests that some of PIP's metabolites may be toxic or have pharmacological potential.

SOCS proteins negatively regulate the immune-related inflammatory responses mediated by the JAK/STAT pathway. Natural product-derived biomolecules targeting JAK/STAT/SOCS signaling have shown potential in treating AD and other inflammatory disorders.

Summary: Previous research has shown that genetic heterogeneity plays a crucial role in modeling amyloid accumulation and other phenotypes seen in Alzheimer's disease. However, the mechanisms behind the impact of genetic background on the pathogenicity of tau aggregates remain unknown. This study induced tau aggregation in mice with different genetic backgrounds and conducted RNA sequencing to identify key genes associated with tau seeding and amyloid response. The findings suggest that the genetic context of mice affects the alteration of transcriptome and tau seeding, providing valuable resources for better modeling tauopathy.

Alkaloids enhanced the cholinergic system, while flavonoids and saponins attenuated neuroinflammation and oxidative stress. These findings suggest that the combination of alkaloid, flavonoid, and saponin fractions derived from FFMN is a promising anti-AD agent.

A study integrated metabolomics and network pharmacology to investigate the effects of Shicao in alleviating acute liver injury

Network pharmacology was used to construct a network of metabolite-reaction-enzyme-gene constituents, and key components and hub genes were screened. Shicao showed hepatoprotective effects against acute liver injury in rats.
