Pentacyclic triterpenoids and ceramide mediate the vasorelaxant activity of Vitex cienkowskii via involvement of NO/cGMP pathway in isolated rat aortic rings

Ethnopharmacological relevance: Vitex cienkowskii Kotschy & Peyritsch is a deciduous tree, prescribed by Cameroonian traditional healers as one of the most popular plant widely used in many disorders including cardiovascular diseases. The preliminary pharmacological studies carried out on Vitex cienkowskii showed its vasorelaxant activities on guinea-pig aortic rings. Aim of the study: The present work evaluated the vasorelaxant activity of extract and isolated compounds from Vitex cienkowskii. Materials and methods: Rat aortic rings were used to evaluate the in vitro vascular effect of the extract. The antioxidant activity was determined by measuring the reduction of the free radical 1,1-diphenyl-1-picryl-hydrazyl (DPPH). Results: Vitex cienkowskii induced significant relaxation in a concentration- and endothelium-dependent manner (EC50 = 12.12 mu g/ml, CH2Cl2-MeOH, 1:1) and did not produce a vasorelaxant effect on contraction evoked by KCl (60 mM). In order to determine its mode of action, Vitex cienkowskii-induced relaxant effect was evaluated in the presence of indomethacin (10 mu M), L-NAME (100 mu M), ODQ (1 mu M) and SQ22356 (100 mu M). Relaxation was significantly blocked by L-NAME and ODQ. These results indicate that Vitex cienkowskii-mediated relaxation is endothelium dependent, probably due to NO release, and the consequent activation of vascular smooth muscle soluble guanylate cyclase (sGC), a signal transduction enzyme that forms the second messenger cGMP. Bio-guided study of Vitex cienkowskii allowed the isolation of the known pentacyclic triterpenoids and a ceramide. It is the first report of salvin A, maslinic acid and a ceramide from Vitex cienkowskii. The activity induced by these compounds indicated that they may be partly responsible for the vasorelaxant effect of the plant extract. A dose of 40 mg/kg of CH2Cl2-MeOH (1:1) extract administered intravenously induced a decrease of mean arterial pressure but did not affect the heart rate. Moreover the plant extracts were found to be highly active in the DPPH radical scavenging assay. Conclusion: Vitex cienkowskii extract possesses antioxidant property, vasorelaxing, and hypotensive effect linked to the endothelium related factors, where nitric oxide is involved. Summary: A new compound, asperpyrone (1), and two known compounds, stigmasterol (2) and 7-hydroxy-3-(2,3-dihydroxybutyl)-1(3H)-isobenzofuranone (3), were isolated from the solid rice culture of the endophytic fungus Aspergillus sp. The structure of asperpyrone (1) was determined through extensive spectral analysis. However, asperpyrone (1) did not show significant amoebicidal activity against Entamoeba moshkovskii in axenic culture.

Summary: The methanol extract of Chrysophyllum lacourtianum stem bark yielded a new secondary metabolite, lacourtianal (1), and eight known compounds. The structures of these compounds were determined by NMR spectroscopic and mass spectrometric analysis. The antibacterial activity of the methanol extract and compounds 1-9 were tested, and lacourtianal (1) and ursolic acid (5) showed strong antibacterial activity.

Summary: The root extract of Nauclea xanthoxylon showed significant inhibition activity against chloroquine resistant and sensitive Plasmodium falciparum strains. Bio-guided fractionation led to the discovery of the new compound xanthoxyloside, which exhibited strong anti-malarial activity. Other compounds with anti-malarial properties were also identified. These findings support the traditional use of Nauclea xanthoxylon in treating malaria.

Summary: Chemical investigation of Gymnanthemum theophrastifolium led to the isolation of a new sesquiterpene (1), a new polyacetylene (2), and a naturally occurring compound (3), along with sixteen known compounds (4-19). Their structures were determined by NMR and MS analysis. This is the first report on the chemical constituents of G. theophrastifolium. The results also provide insights into the chemophenetic knowledge of G. theophrastifolium.

Summary: The phytochemical investigation of Helichrysum foetidum led to the isolation of four undescribed compounds, including two ent-beyer-15-ene-type diterpenoids, foetidumins A and B, one flavonoid, foetidumin C, and one chalcopyrone, foetidumin D. In addition, fourteen known compounds were also isolated. Foetidumin C, erythroxylol A, and kaempferol showed the highest antileishmanial potency. The extracts of flowers and twigs also exhibited high activity against Plasmodium falciparum.

Summary: A new unsaturated aliphatic anhydride derivative and ten known compounds were isolated from a fungus inhabiting Pittosporum mannii. The p-terphenyls are suggested as the chemophenetic marker for the genus Aspergillus. The tested sample showed good antibiofilm activity, low anti-inflammatory activity, and moderate cytotoxicity.

The antibacterial and antiplasmodial activities of the extracts and isolated compounds were evaluated. The fraction E showed significant activity against Staphylococcus aureus, while certain compounds exhibited high activity against Plasmodium falciparum 3D7 strain.

Summary: This study aimed to identify the cytotoxic constituents of Brucea antidysenterica and demonstrate their mode of induction of apoptosis on cancer cell lines. The results showed that Brucea antidysenterica and its constituents have potential antiproliferative effects and can induce apoptosis through caspase activation, alteration of mitochondrial membrane potential, and increased reactive oxygen species levels.

Co-Immobilization of a Multi-Enzyme Cascade: (S)-Selective Amine Transaminases, l-Amino Acid Oxidase and Catalase

Summary: An enzyme cascade consisting of l-amino acid oxidase (hcLAAO4) and catalase (hCAT) was immobilized together with the (S)-selective amine transaminase (ATA-Vfl) to achieve higher reaction rates and more efficient H2O2-removal. The co-immobilized enzyme cascade showed good stability and high enantiomeric purity in preparative kinetic resolutions. However, further recycling was limited due to the instability of ATA-Vfl. The use of an engineered ATA-Vfl-8M in the immobilized enzyme cascade allowed a significant reduction in the amount of co-substrate required.

Extended Biocatalytic Halogenation Cascades Involving a Single-Polypeptide Regeneration System for Diffusible FADH(2)

Summary: In this study, a bifunctional regeneration enzyme for two-step catalytic flavin regeneration was introduced, using phosphite as an inexpensive sacrificial substrate. This fusion protein showed good performance when co-expressed with various flavin-dependent Trp-halogenases, enabling carrier-free immobilization for protein production and the preparative synthesis of halotryptophan. The scalability of this system was demonstrated in bench-top bioreactors on a 2.5 L scale. Furthermore, the inclusion of a 6-halotryptophan-specific dioxygenase into the co-expression strain allowed for the conversion of the halogenation product to the kynurenine derivative, enabling the one-pot synthesis of L-4-Cl-kynurenine and its brominated analogue on a preparative scale.

Summary: Due to the increasing antibiotic resistance, there is an urgent need to search for antimicrobial substances that can disrupt bacterial biofilms and quorum sensing. In this study, twenty compounds isolated from the medicinal plant Gambeya lacourtiana were evaluated for their antibiofilm and anti-quorum sensing effects against selected pathogenic bacteria. The results showed that most of the compounds inhibited violacein production and disrupted signal molecule reception.

Ameliorative effect of scopolamine-induced cognitive dysfunction by Fufangmuniziqi formula: The roles of alkaloids, saponins, and flavonoids

Summary: This study investigated the effects of Fufangmuniziqi formula (FFMN) on Alzheimer's disease (AD) and the role of alkaloids, flavonoids, and saponins in the process. The results showed that FFMN improved cognitive and memory impairment in the AD mouse model. Alkaloids enhanced the cholinergic system, while flavonoids and saponins attenuated neuroinflammation and oxidative stress. These findings suggest that the combination of alkaloid, flavonoid, and saponin fractions derived from FFMN is a promising anti-AD agent.

The results showed that M. longifolia extract is rich in phenolic compounds and has no observed toxicity up to a certain dosage limit.

A study integrated metabolomics and network pharmacology to investigate the effects of Shicao in alleviating acute liver injury

Summary: This study aimed to explore the hepatoprotective effects of Shicao on CCl4-induced acute liver injury. A rat model of acute liver injury was established and liver function indices were assessed. Untargeted metabolomics was conducted to identify differential metabolites. Network pharmacology was used to construct a network of metabolite-reaction-enzyme-gene constituents, and key components and hub genes were screened. Shicao showed hepatoprotective effects against acute liver injury in rats.
