Berberine and Coptidis Rhizoma as novel antineoplastic agents: A review of traditional use and biomedical investigations

Ethnopharmacological relevance: Coptidis Rhizoma (Huanglian) and its major component, berberine, have drawn extensive attention toward their antineoplastic effects in the recent years. The antineoplastic effects are related to the Chinese Medicine (CM) properties of Huangliang in treating diseases by removing damp-heat and purging fire and Counteracting toxicity. Aim of the review: To trace the long history of the traditional use of Huanglian from folk medicines, especially from Chinese medicine, to recent pharmacological studies of Huanglian and berberine, with all emphasis on their antineoplastic effects and the promise as novel a antineoplastic agents. Methods: A total of seven databases were extensively searched for literature research. The terms and keywords for Searching included Huanglian, berberine, Coptis, Coptidis Rhizoma, anticancer, anti-invasion, antimatastasis and mechanism. The papers including Ours With studies oil anticancer and mechanism, pharmacology and toxicology of Huanglian and/or berberine were focused. Results: In view of traditional use, the anticancer effects of Huanglian can be ascribed to its CM trait by removing damp-heat,fire and toxicity. From modern biomedical Studies, anticancer effects have been demonstrated in both Huanglian and berberine. The underlying molecular mechanisms involve cell-cycle arrest, apoptosis induction and anti-inflammation. Berberine is an essential anticancer compound in Huanglian. in Some Studies, the use of Huanglian was shown to be more effective and beneficial than the use of berberine alone. The presence of other protoberberine-type alkaloids in Huanglian might give synergistic effects for the anticancer effects. Berberine also demonstrates effects of antiangiogenesis, anti-invasion and anti-metastasis in some cancer cell lines. however, more investigations are required to unravel the underlying mechanisms involved. Conclusions: The modern evidences of treating cancer with Huanglian and berberine have a strong linkage with traditional concept and rules Of using Huanglian in CM practice. As anticancer candidates with low toxicity. berberine and its altered structure, as well as Huanglian and its formulae, will attract scientists to pursue the potential anticancer effects and the mechanisms by using technologies of genomics, proteomics and other advanced approaches. Oil the other hand. relatively few in vivo studies have been conducted oil anticancer effects of Huanglian and berberine. The clinical application of berberine or Huanglian as novel cancer therapeutic agents requires in vivo validations and further investigations of their anticancer mechanisms. Recent research shows that the postoperative tumor microenvironment (TME) plays a crucial role in tumor recurrence. This review discusses the impact of surgical stress and perturbation on the TME and highlights the potential of targeting the postoperative TME for adjuvant therapeutics.

Deciphering suppressive effects of Lianhua Qingwen Capsule on COVID-19 and synergistic effects of its major botanical drug pairs

Summary: The COVID-19 pandemic has led to excessive deaths globally. Conventional antiviral drugs have limited effectiveness in relieving symptoms, but Lianhua Qingwen Capsule has shown remarkable anti-COVID-19 effects. This review aims to uncover the pharmacological actions of Lianhua Qingwen Capsule, verify its bioactive ingredients, investigate the compatibility of botanical drug pairs, and clarify the clinical evidence and safety of combined therapy with conventional drugs. Several bioactive ingredients in Lianhua Qingwen Capsule were found to target host cytokines and regulate immune defense against COVID-19. Genes such as androgen receptor, myeloperoxidase, epidermal growth factor receptor, insulin, and aryl hydrocarbon receptor were significantly involved in its pharmacological actions. Four botanical drug pairs in Lianhua Qingwen Capsule exhibited a synergistic effect in treating COVID-19. Clinical studies demonstrated the medicinal effect of combining Lianhua Qingwen Capsule with conventional drugs against COVID-19. In conclusion, the main pharmacological mechanisms of Lianhua Qingwen Capsule for managing COVID-19 have been revealed, showing therapeutic effects against the disease.

Ameliorative effect of scopolamine-induced cognitive dysfunction by Fufangmuniziqi formula: The roles of alkaloids, saponins, and flavonoids

Summary: This study investigated the effects of Fufangmuniziqi formula (FFMN) on Alzheimer's disease (AD) and the role of alkaloids, flavonoids, and saponins in the process. The results showed that FFMN improved cognitive and memory impairment in the AD mouse model. Alkaloids enhanced the cholinergic system, while flavonoids and saponins attenuated neuroinflammation and oxidative stress. These findings suggest that the combination of alkaloid, flavonoid, and saponin fractions derived from FFMN is a promising anti-AD agent.

This study analyzed the phytochemical profile of the aqueous extract of M. longifolia and evaluated its acute and subacute toxicity in mice. The results showed that M. longifolia extract is rich in phenolic compounds and has no observed toxicity up to a certain dosage limit.

A study integrated metabolomics and network pharmacology to investigate the effects of Shicao in alleviating acute liver injury

Summary: This study aimed to explore the hepatoprotective effects of Shicao on CCl4-induced acute liver injury. A rat model of acute liver injury was established and liver function indices were assessed. Untargeted metabolomics was conducted to identify differential metabolites. Network pharmacology was used to construct a network of metabolite-reaction-enzyme-gene constituents, and key components and hub genes were screened. Shicao showed hepatoprotective effects against acute liver injury in rats.
