Summary: This study reports a new method for the rapid synthesis of 2-deoxy-3-thio pyranoses and their O-glycosides through the reaction of glycals with two different types of nucleophiles in the presence of SnCl4. The reaction involves thioaryl substitution at C-3 and alpha-selective O-glycosylation at C-1, combining two reactions with three interventions. The method offers operational simplicity at 0 degrees C in 10-20 minutes and is compatible with a wide range of primary and secondary alcohols, yielding products in good to excellent yields. Moreover, the strategy has the potential for the synthesis of various natural products and drug molecules, as supported by control experiments.

The constructed dual-site ADCs showed acceptable homogeneity, excellent buffer stability, and enhanced in vitro and in vivo efficiency.

Summary: This study reports a one-pot strategy for functionalizing pyranoside 1,2-cis-diols with two different ester protecting groups. The approach selectively acylates through orthoester hydrolysis promoted by a carboxylic acid, and after removal of water and introduction of a coupling agent, the carboxylic acid is esterified with the liberated hydroxyl group. Although applied to 1,2-cis-diols on pyranoside scaffolds, this method can be applicable to similar motifs on any six-membered ring.

Summary: This study presents a one-pot strategy for stereocontrolled glycosylation to generate both alpha- and beta-glycosides by altering reaction conditions. The selective formation of beta-glycosides was achieved by activating the perbenzylated trichloroacetimidate with a catalytic amount of SnCl4 at -40 degrees C. On the other hand, alpha-glycosides were obtained as the major products at room temperature by using 3.0 equivalents of SnCl4 and/or TiCl4. Control experiments and DFT calculations provided mechanistic insights into the glycosylation reaction, revealing a sequential occurrence of beta-glycosylation and anomerization.

Techniques like OSMAC, overexpression/deletion of regulatory genes, ribosome engineering, and promoter replacement have been utilized to enhance the production titer of target compounds. Heterologous expression of BGCs has successfully been employed for the detection, characterization, and production of many secondary metabolites.

Summary: In this study, a sugar-O-methyltransferase ThnM1 was identified and functionally characterized. The enzyme showed promiscuity and regiospecific activity at the 2'-OH of rhamnose. By using engineered Escherichia coli, a methoxy-rhamnosyl derivative of quinizarin was successfully produced. The cytotoxicity of the derivative was studied and compared with that of quinizarin. This study provides insights into the potential diversification of natural products through the methylation of the rhamnose moiety.

Summary: Anthraquinone and its derivatives have remarkable biological properties, such as anticancer, antibacterial, antifungal, and antiviral activities. This study developed a recombinant Escherichia coli strain to modify chrysazin to chrysazin-8-O-alpha-l-rhamnoside (CR) and chrysazin-8-O-alpha-l-2'-O-methylrhamnoside (CRM), which showed higher antibacterial and anticancer properties than chrysazin.

Summary: In this study, a biosynthetic gene cluster (BGC) for Lyngbyastatins (Lbns) was identified and characterized in a marine cyanobacterium. The BGC was found to encode enzymes responsible for the production of compounds 1 and 2, which have anti-cancer activity. Similar BGCs were also identified in the genomes of other marine cyanobacteria.

Summary: A subcluster of cytotoxic apratoxin A was isolated from an environmental DNA library and engineered for expression in Anabaena sp. PCC7120. By further engineering with a rhamnose-inducible promoter, a chiral building block necessary for the synthesis of apratoxin analogues was successfully produced, marking the first synthetic biology attempt for this type of polyketide fragment.

Summary: By activating the new antibacterial compound peucemycin using an OSMAC strategy, researchers discovered that it is a novel gamma-pyrone macrolide. Peucemycin was successfully activated at 18 degrees Celsius, showing potential antibacterial activity and a suppressive effect on the viability of cancer cell lines.
