Enhanced Production of Nargenicin A(1) and Generation of Novel Glycosylated Derivatives

Nargenicin A(1), an antibacterial polyketide macrolide produced by Nocardia sp. CS682, was enhanced by increasing the pool of precursors using different sources. Furthermore, by using engineered strain Nocardia sp. ACC18 and supplementation of glucose and glycerol, enhancement was similar to 7.1 fold in comparison to Nocardia sp. CS682 without supplementation of any precursors. The overproduced compound was validated by mass spectrometry and nuclear magnetic resonance analyses. The novel glycosylated derivatives of purified nargenicin A(1) were generated by efficient one-pot reaction systems in which the syntheses of uridine diphosphate (UDP)-alpha-D-glucose and UDP-alpha-D-2-deoxyglucose were modified and combined with glycosyltransferase (GT) from Bacillus licheniformis. Nargenicin A(1) 11-O-beta- D-glucopyranoside, nargenicin A(1) 18-O-beta-D-glucopyranoside, nargenicin A(1)11 18-O-beta-D- diglucopyranoside, and nargenicin 11-O-beta-D-2-deoxyglucopyranoside were generated. Nargenicin A(1) 11-O-beta-D-glucopyranoside was structurally elucidated by ultra-high performance liquid chromatography-photodiode array (UPLC-PDA) conjugated with high-resolution quantitative time-of-flight-electrospray ionization mass spectroscopy (HR-QTOF ESI-MS/MS), supported by one- and two-dimensional nuclear magnetic resonance studies, whereas other nargenicin A(1) glycosides were characterized by UPLC-PDA and HR-QTOF ESI-MS/MS analyses. The overall conversion studies indicated that the one-pot synthesis system is a highly efficient strategy for production of glycosylated derivatives of compounds like macrolides as well. Furthermore, assessment of solubility indicated that there was enhanced solubility in the case of glycoside, although a substantial increase in activity was not observed.

Recent Advances in the Heterologous Biosynthesis of Natural Products from Streptomyces

Summary: Streptomyces is an important source of natural products used in therapeutic antibiotics, anticancer agents, pesticides, and dyes. Advances in metabolite analysis have led to the characterization of many new secondary metabolites. Techniques like OSMAC, overexpression/deletion of regulatory genes, ribosome engineering, and promoter replacement have been utilized to enhance the production titer of target compounds. Heterologous expression of BGCs has successfully been employed for the detection, characterization, and production of many secondary metabolites.

Functional Characterization of a Regiospecific Sugar-O-Methyltransferase from Nocardia

Summary: In this study, a sugar-O-methyltransferase ThnM1 was identified and functionally characterized. The enzyme showed promiscuity and regiospecific activity at the 2'-OH of rhamnose. By using engineered Escherichia coli, a methoxy-rhamnosyl derivative of quinizarin was successfully produced. The cytotoxicity of the derivative was studied and compared with that of quinizarin. This study provides insights into the potential diversification of natural products through the methylation of the rhamnose moiety.

Microbial Biosynthesis of Chrysazin Derivatives in Recombinant Escherichia coli and Their Biological Activities

Summary: Anthraquinone and its derivatives have remarkable biological properties, such as anticancer, antibacterial, antifungal, and antiviral activities. This study developed a recombinant Escherichia coli strain to modify chrysazin to chrysazin-8-O-alpha-l-rhamnoside (CR) and chrysazin-8-O-alpha-l-2'-O-methylrhamnoside (CRM), which showed higher antibacterial and anticancer properties than chrysazin.

Biosynthesis of Lyngbyastatins 1 and 3, Cytotoxic Depsipeptides from an Okeania sp. Marine Cyanobacterium

Summary: In this study, a biosynthetic gene cluster (BGC) for Lyngbyastatins (Lbns) was identified and characterized in a marine cyanobacterium. The BGC was found to encode enzymes responsible for the production of compounds 1 and 2, which have anti-cancer activity. Similar BGCs were also identified in the genomes of other marine cyanobacteria.

Heterologous Production of the C33-C45 Polyketide Fragment of Anticancer Apratoxins in a Cyanobacterial Host

Summary: A subcluster of cytotoxic apratoxin A was isolated from an environmental DNA library and engineered for expression in Anabaena sp. PCC7120. By further engineering with a rhamnose-inducible promoter, a chiral building block necessary for the synthesis of apratoxin analogues was successfully produced, marking the first synthetic biology attempt for this type of polyketide fragment.

Identification and enhancing production of a novel macrolide compound in engineered Streptomyces peucetius

Summary: By activating the new antibacterial compound peucemycin using an OSMAC strategy, researchers discovered that it is a novel gamma-pyrone macrolide. Peucemycin was successfully activated at 18 degrees Celsius, showing potential antibacterial activity and a suppressive effect on the viability of cancer cell lines.
