Lipid- and polymer-based cationic nanoparticles are utilized for different types of cancer vaccines, and their potential in T-cell induction is discussed for therapeutic cancer vaccination.

Summary: Cationic liposome delivery of interfering RNA (shRNA) modified with cyclic Arg-Gly-Asp (cRGD) shows promising targeted delivery of ONECUT2 (OC-2) shRNA to breast cancer cells. The cRGD-CL/pshOC-2 lipoplexes have uniform particle size, moderate zeta potential, and high encapsulation efficiency. The pshOC-2 down-regulates OC-2 expression and exhibits antitumor effects. The intravenous administration of cRGD-CL/pshOC-2 lipoplexes reduces OC-2 expression in tumors and shows significant antitumor effects.

Summary: Effective interaction with biomembranes is crucial for the activity of photosensitizers. In this study, amphiphilic cationic phthalocyanines with different bulkiness of substitution on the lipophilic part (-H, -SMe, -StBu) were prepared and their interaction with biomembranes was investigated. The results showed that even the bulky -StBu derivatives could enter biomembranes and exhibited good photosensitizing activity. Moreover, these derivatives demonstrated excellent anti-tumor activity in various cell lines.

Summary: Engineered cytokine variants of IL-12 and IL-2 represent a rational approach to combination cytokine therapy for cancer immunotherapy. The combination of IL-12 and IL-2 enhances the effector differentiation of tumor-reactive CD8+ T cells, decreases the number of tumor-infiltrating CD4+ regulatory T cells, and prolongs the survival of lung tumor-bearing mice.

Summary: The study investigates the influence of hydrophilic surfactants on the membrane elasticity of liposomes and their effect on the skin absorption of vitamin C. Encapsulation of vitamin C inside cationic liposomes is aimed at improving its delivery to the skin. Elastic liposomes (ELs) formed by the addition of Polysorbate 80 to conventional liposomes (CLs) were compared. The presence of a positive charge in the liposomal membrane significantly increases the encapsulation efficacy of both CLs and ELs, indicating a potential mechanism involving interactions between cationic lipids and vitamin C during skin diffusion.

Summary: The study focused on the development of cationic fusogenic liposomes with intercalated radioactive [I-131]I- for potential therapeutic applications. Experiment results showed that these liposomes, with high membrane fusion ability, had a special delivery pathway and exhibited excellent delivery efficiency in in vitro experiments, suitable for invasive cancer cell models.

Summary: This study assessed the efficacy of cationic lipid-based systems for the delivery of HER2/neu siRNA in breast cancer. The results showed that cationic liposomes efficiently bound, compacted, and protected the therapeutic siRNA against degradation. Summary: The tumor microenvironment (TME) is a complex mixture of resident and tumor cells that communicate closely through secretory products. While immune cells play a significant role in eliminating tumor cells, they can also contribute to tumor development in certain cases. In cervical cancer (CC), the human papilloma virus (HPV) shapes the TME to facilitate persistent infections and promote tumor growth. Interleukin-2 (IL-2) is an important cytokine in the TME that induces the elimination of tumor cells by CD8+ effector T cells and natural killer cells, but it can also suppress immune responses through Treg cells. Recent studies have revealed that CC cells express the IL-2 receptor (IL-2R) and can proliferate in response to low concentrations of exogenous IL-2, potentially depleting this molecule to evade immune responses. This review provides an overview of the immune cells in the TME of CC and the involvement of IL-2 in tumor promotion.

Summary: The study found that encapsulating paclitaxel in large cationic liposomes can avoid peripheral neurotoxicity and improve the drug's anti-tumor effects on lung cancer. These liposomes not only show good biocompatibility, but also more effectively target cancer stem cells and tumors, reducing the painful peripheral neuropathy caused by the drug.

Summary: In this study, cationic nanogels were incorporated into liposomes for stable and enhanced mRNA delivery to cells. The use of DNPR9 with liposomes resulted in enhanced internalization and a 4-fold increase in transfection efficiency compared to control liposomes. The condensation strategy using DNPR9 can be widely applied to enhance the efficacy of various lipid-based formulations.

Summary: In response to the global need for an effective vaccine in the SARS-CoV-2 pandemic, a plasmid DNA vaccine expressing the spike (S) protein was developed and tested for in vitro transfection and in vivo immunogenicity. The formulated liposomes complexed with the DNA vaccine showed enhanced neutralizing antibody responses and T cell responses, comparable to the efficacy of intramuscular electroporation (IM-EP) as a benchmark.
