Summary: Riboswitches are known to modulate transcription elongation or translation initiation, but a study on guanidine III riboswitch reveals that ligand binding can also protect mRNA from degradation by creating a structure that impedes scanning by regulatory endonuclease RNase E. This finding suggests that riboswitch aptamers can independently regulate gene expression by prolonging mRNA longevity in response to ligand binding.

Summary: This article reveals the intricate post-transcriptional networks mediated by GlmZ in the biogenesis of GlcN6P, an essential precursor of the bacterial cell envelope. The cryoEM structure of the RapZ:GlmZ complex is presented, showing a complementary match between the RapZ tetrameric quaternary structure and the structural repeats in the sRNA. The structure of a precleavage intermediate formed between the binary RapZ:GlmZ complex and RNase E is also presented, illustrating how GlmZ is recognized by the enzyme.

Summary: In enteric bacteria, small RNAs (sRNAs) such as MicC stimulate target RNA decay using RNase E. The interaction between the sRNA 5' monophosphate (5 ' P) and RNase E activates cleavage of the target. However, experimental evidence supporting this model is limited. To address this, researchers developed a genetic tool to generate controllable 5' monophosphorylated sRNAs. The results showed that while the 5' phosphorylation state determines stability of naturally triphosphorylated sRNAs, it does not play a major role in target RNA destabilization in vivo. Additionally, the C-terminal half of RNase E is crucial for MicC-mediated decay, suggesting the mandatory interaction with Hfq.
