Detection of hair-microRNAs as the novel potent biomarker: Evaluation of the usefulness for the diagnosis of scleroderma

Background: Serum microRNA levels are known as useful biomarkers for various diseases. Recent publication has indicated the existence of microRNAs in hair roots and hair shafts. Objective: In this study, we evaluated several methods for the extraction of hair microRNAs, and their usefulness for the diagnosis of scleroderma. Methods: A single hair root and 5 pieces of hair shafts were obtained from the occiput of each individual of 11 scleroderma patients and 13 normal subjects at the time of serum sampling. microRNA extraction from sera or hair roots was performed with commercially available kits. microRNAs were extracted from hair shafts using four different methods. microRNA expression was evaluated by PCR array and real-time PCR. Results: We demonstrated microRNAs in hair roots and hair shafts were detectable and quantitative using our method. We found the difference of microRNA levels in hair roots and hair shafts obtained from different places of head in each individual were within 2-fold, indicating the reproducibility of hair microRNA levels by our method. PCR array revealed microRNAs from sera, hair roots and hair shafts have different expression pattern, and can be independent biomarkers. Serum and hair root miR-196a levels were not significantly changed in scleroderma patients, while we found miR-196a levels in hair shafts were significantly decreased in scleroderma patients compared to those in normal subjects (p < 0.05). Conclusion: Hairs are more accessible than sera among human samples. microRNAs levels in hair roots or hair shafts may become effective and independent biomarkers. Bioactive substances in the stratum corneum of the epidermis found as indicators of skin damage due to sun exposure

Summary: The study investigated the potential of bioactive substances in the stratum corneum as early and sensitive indicators of sun exposure on the skin using ROC analysis. Results showed that IL-1ra/IL-1 alpha, TNF-alpha, and Bax in the stratum corneum are sensitive to sun exposure and decreased significantly after sunscreen application, indicating their potential as markers for sun damage.

Promising Blood-Based Biomarkers for Melanoma: Recent Progress of Liquid Biopsy and Its Future Perspectives

Summary: Because of the success of novel therapeutic approaches, less invasive and repeatable monitoring tools are needed for melanoma. Liquid biopsy, specifically microRNA, cell-free DNA, and circulating tumor cells obtained from blood samples, has shown promising potential as a diagnostic and prognostic tool for melanoma. Despite challenges, recent technical advances have improved the sensitivity and provided high-throughput analysis for liquid biopsy. Standardizing assay procedures and increasing sensitivity are necessary for widespread clinical implementation, but liquid biopsy could be utilized quickly. This review emphasizes the importance of liquid biopsy and its potential future perspectives.

The patients were successfully treated with fosravuconazole and terbinafine, respectively. A summary of 32 cases of cutaneous and subcutaneous phaeohyphomycosis caused by Exophiala species in Japan was included, with different antifungal treatments and their success rates discussed.

Summary: Cancer immunotherapy is the primary treatment for unresectable cancers, but it is not a complete cure for all patients. Immune cell therapy, specifically using immune cells generated from induced pluripotent stem cells (iPSCs), shows promise for innovative cancer immunotherapy methods. iPSC-derived dendritic cells (iPS-DCs) can activate T cells, while iPSC-derived macrophages (iPS-MPs) attack cancer cells. iPSCs offer a source for genetic modification and addition of immune functions. Additionally, immortalized iPS-DCs and iPS-MPs could potentially reduce costs through mass production. This review summarizes the achievements and future prospects of cancer research using iPS-DCs and iPS-MPs.

Summary: Xeroderma pigmentosum (XP) is a rare genodermatosis characterized by photosensitivity and a high risk of skin tumors due to DNA repair deficiency. This study identified two deep intronic mutations in the ERCC4/XPF gene in 17 cases of XP-F, a rare subtype of XP. These mutations result in reduced gene expression and early-onset skin cancers, highlighting the need for attention to these variants. Additionally, antisense oligonucleotides designed for these mutations can restore DNA repair capacity, suggesting potential therapeutic targets for XP-F.
