Summary: Traditionally, en coup de sabre form of morphea involving the scalp often results in sclerosis, dyspigmentation, and scarring alopecia that is thought to be permanent and unresponsive to treatment. However, this report presents two cases where extensive scarring alopecia improved with medical treatment.

Summary: This study demonstrates that DLX5 promotes hair follicle stem cell (HFSC) differentiation by modulating the c-MYC/miR-29c-3p/NSD1 axis. DLX5 is highly expressed in differentiated HFSCs and transcriptionally activates c-MYC expression to induce HFSC differentiation. c-MYC suppresses miR-29c-3p expression, leading to NSD1-mediated HFSC differentiation.

Summary: This study investigated the expression profiles of miRNA and mRNA in cashmere goat fetal skin, and identified miRNAs and mRNAs related to the development of secondary hair follicles. Enrichment analysis revealed that some miRNA target genes were consistent with the screened mRNAs and enriched in Notch and TGF-beta signaling pathways. A regulatory network for the development of secondary hair follicles was constructed, and a targeted relationship between miR-30e-5p and DLL4 was confirmed. This study provides insights into the molecular mechanism of miRNA-mRNA in the development of hair follicles in cashmere goats.

Summary: By analyzing clinical characteristics and laboratory values, we identified strategies to improve trial design for early diffuse SSc, including skin thickness progression rate, RNA polymerase III antibody positivity, and presence of tendon friction rubs. Applying these strategies to actual trial data can reduce the decline in placebo group skin scores and reveal significant differences in the CRISS score.

Summary: Regardless of whether the first SSc manifestation or first non-Raynaud manifestation is used to define disease onset, a disease duration of less than 18 months at enrollment is preferable. A longer disease duration criterion more frequently results in regression to the mean of the mRSS score, which likely contributes to negative trial outcomes.

Summary: Human hair is increasingly used as a noninvasive matrix for assessing human exposure to organic contaminants. This study investigated the accumulation processes of organic contaminants in hair by examining changes in hair length and color in a Chinese population. The results showed that polychlorinated biphenyls and organophosphate flame retardants increased along the hair shaft with hair growth and distance from the scalp. The levels of organic contaminants also decreased with the change from black to white hair, likely due to the loss of melanin.

Summary: The study assessed DNA methylation differences between juvenile systemic sclerosis (jSSc) and juvenile localized scleroderma (jLS), revealing distinct epigenetic patterns and unique methylated genes in both diseases. Compared to healthy controls, differential methylation sites and genes were identified in jSSc and jLS, indicating potential novel diagnostic biomarkers for localized scleroderma. Pathway analysis showed enrichment of inflammatory pathways in jSSc and the HIPPO signaling pathway in jLS, highlighting different biological processes involved in each condition.

Summary: The study investigated the potential of bioactive substances in the stratum corneum as early and sensitive indicators of sun exposure on the skin using ROC analysis. Results showed that IL-1ra/IL-1 alpha, TNF-alpha, and Bax in the stratum corneum are sensitive to sun exposure and decreased significantly after sunscreen application, indicating their potential as markers for sun damage.

Summary: The study found that EV miR-92a-2-5p levels in serum were negatively correlated with adverse reactions after COVID-19 vaccination, while EV miR-148a levels were associated with specific antibody titers. This suggests that circulating EV miRNAs could serve as biomarkers for vaccine efficacy and adverse reactions.

Summary: Because of the success of novel therapeutic approaches, less invasive and repeatable monitoring tools are needed for melanoma. Liquid biopsy, specifically microRNA, cell-free DNA, and circulating tumor cells obtained from blood samples, has shown promising potential as a diagnostic and prognostic tool for melanoma. Despite challenges, recent technical advances have improved the sensitivity and provided high-throughput analysis for liquid biopsy. Standardizing assay procedures and increasing sensitivity are necessary for widespread clinical implementation, but liquid biopsy could be utilized quickly. This review emphasizes the importance of liquid biopsy and its potential future perspectives.

Summary: Cancer immunotherapy is the primary treatment for unresectable cancers, but it is not a complete cure for all patients. Immune cell therapy, specifically using immune cells generated from induced pluripotent stem cells (iPSCs), shows promise for innovative cancer immunotherapy methods. iPSC-derived dendritic cells (iPS-DCs) can activate T cells, while iPSC-derived macrophages (iPS-MPs) attack cancer cells. iPSCs offer a source for genetic modification and addition of immune functions. Additionally, immortalized iPS-DCs and iPS-MPs could potentially reduce costs through mass production. This review summarizes the achievements and future prospects of cancer research using iPS-DCs and iPS-MPs.

Summary: Xeroderma pigmentosum (XP) is a rare genodermatosis characterized by photosensitivity and a high risk of skin tumors due to DNA repair deficiency. This study identified two deep intronic mutations in the ERCC4/XPF gene in 17 cases of XP-F, a rare subtype of XP. These mutations result in reduced gene expression and early-onset skin cancers, highlighting the need for attention to these variants. Additionally, antisense oligonucleotides designed for these mutations can restore DNA repair capacity, suggesting potential therapeutic targets for XP-F.
