Nuclear IL-33 regulates cytokinesis and cell motility in normal human epidermal keratinocytes

Hidetoshi Tsuda, Shin-ichi Tominaga, Mamitaro Ohtsuki, Mayumi Komine

Summary: This study investigated the role of IL-33 in normal human epidermal keratinocytes (NHEKs) by utilizing RNA interference to knock down cellular IL-33. The results showed that IL-33 knockdown affected cell division and motility, possibly by attenuating ECT2 expression and slightly decreasing RhoA activity.

Summary: In recent years, circulating cell-free DNA (cfDNA) has gained attention as a biomarker for cancer. This study found that serum cfDNA levels in patients with cutaneous T-cell lymphoma (CTCL) were significantly higher than healthy controls and increased with disease progression. There were positive correlations between cfDNA levels and certain blood markers and clinical findings, and higher cfDNA levels were associated with a worse prognosis in CTCL patients.

Inflammation Causes Exacerbation of COVID-19: How about Skin Inflammation?

Mayumi Komine, Tuba Mussarat Ansary, Md Razib Hossain, Koji Kamiya, Mamitaro Ohtsuki

Summary: COVID-19 is a global viral infection caused by SARS-CoV-2, possibly originating from bat coronaviruses in China. Deforestation by humans may have led to host conversion and exposure of wild bats to new environments. SARS-CoV-2 infects cells through two mechanisms, and obesity and diseases like hypertension, diabetes, and pulmonary diseases can result in poor prognosis of COVID-19.

Comparison of transcriptomic profiles in edge to center of plaque reveals chronological molecular events in psoriatic plaque formation

Suphagan Boonpethkaew, Jitlada Meephansan, Onjira Jumlongpim, Sasin Charoensuksira, Pattarin Tangtanatakul, Jongkonnee Wongpiyabovorn, Mayumi Komine

Summary: This study compared the gene profiles in the peripheral edge (PE) and center (CE) of plaques to elucidate the chronological molecular events in plaque formation. Inflammation is likely initiated in the PE, characterized by the presence of specific molecules such as IL-36 alpha, S100A7, and S100A15. The inflammatory state progresses towards a more stable state in the CE, where growth factor activities are increased, potentially leading to the regression of inflammation. These findings contribute to the development of novel therapies for psoriasis.

Epigenetic control of skin immunity

Sayaka Shibata

Summary: Epigenetics plays a crucial role in immune cell activation in the skin. Disruption of normal epigenetic regulation can lead to dysregulation of immune factors and impact skin immunity. This review focuses on the epigenetic regulation of histone modification and chromatin remodeling and their potential involvement in chronic cutaneous immune disorders.

Impact of Pretreatment Systemic Inflammatory Markers on Treatment Persistence with Biologics and Conventional Systemic Therapy: A Retrospective Study of Patients with Psoriasis Vulgaris and Psoriatic Arthritis

Eiki Sugimoto, Hiroki Matsuda, Sayaka Shibata, Yuka Mizuno, Asumi Koyama, Lixin Li, Haruka Taira, Yukiko Ito, Kentaro Awaji, Takashi Yamashita, Shinichi Sato

Summary: This study examined accessible systemic inflammatory markers in patients with psoriasis vulgaris and psoriatic arthritis, and evaluated their association with psoriasis severity, the presence of arthritis, and treatment continuation rates. The findings showed positive correlations between various inflammatory markers and Psoriasis Area and Severity Index scores. Higher platelet/lymphocyte ratio (PLR) or CRP values were associated with psoriatic arthritis diagnosis. Higher pretreatment neutrophil or platelet count, PLR, and SII were associated with lower treatment continuation rates of conventional systemic agents. These findings suggest that accessible inflammatory markers can effectively assess systemic inflammation and guide therapeutic approaches for psoriasis vulgaris and psoriatic arthritis.
