Summary: This study compared the outcome of facial symmetry in patients with craniofacial malformations who underwent simultaneous digitally planned patient-specific implant (PSI-) based orthognathic surgery and polyether ether ketone (PEEK) bone augmentation. The results showed that simultaneous PEEK bone augmentation significantly improves facial symmetry compared to conventional orthognathic surgery, even in patients with syndrome-related underdevelopment.

Summary: Conventional histology is based on evaluating 2D slices of a 3D biopsy, while 3D X-ray histology overcomes this limitation by enhancing soft tissue X-ray contrast through tissue staining before CT imaging. The ideal X-ray contrast agent, dibromo fluorescein barium salt, allows for 3D imaging and subsequent complementing counterstaining using standard histological techniques.

Summary: This study investigated the developmental toxicity of Butylparaben (BuP) in zebrafish embryos, and found that exposure to BuP caused severe craniofacial deformities, periocular edema, cardiac dysplasia, and delayed otolith development in zebrafish larvae. The oxidative stress response was enhanced, and the activities of catalase and superoxide dismutase were reduced while the concentration of malondialdehyde was elevated. In addition, ALP activity and the expression of chondrocyte marker genes were decreased. These findings indicate that BuP-induced oxidative stress inhibits the proliferation of neural crest cells (NCCs), leading to craniofacial deformities.

Summary: Concrete structures produced by extrusion-based 3D printing exhibit orthotropic mechanical behavior and relative weakness in interfacial regions. The microstructure, including porosity content and distribution, plays a key role in these properties and can be visualized and quantified using X-ray computed tomography. Further research is needed to understand the effect of stress concentrations at micro-cracks or voids in 3D printed concrete on mechanical behavior.

Summary: This article introduces the important roles of various signaling mediators in the Hh family in different stages of development, and the significance of the Hh pathway in bone tissue development and tumorigenesis. Gorlin syndrome is a skeletal and tumorigenic disorder caused by gain-of-function mutations in Hh signaling. The article first presents the phenotype of Gorlin syndrome and the relationship between genotype and phenotype in bone and craniofacial tissues, including the causative gene as well as other Hh-related genes. Next, the importance of new diagnostic methods using next-generation sequencing and multiple gene panels is discussed. Finally, the article summarizes Hh-related genetic disorders, including cilia disease, and the genetics of Hh-related bone diseases.
